摘 要:目的:探讨谷氨酰胺对热应激雌鸡生殖机制的影响。方法:选用90只健康的14日龄雌性雏鸡,预饲7 d后,随机分为5个组,每组18只:Ⅰ组为对照组(常温),饲喂基础日粮;Ⅱ~Ⅴ组为热应激试验组,分别在基础日粮中添加0%、0.5%、1.0%、1.5%谷氨酰胺。试验持续21 d,分别在28、35、42日龄随机选取3只鸡,屠宰后采集血液和卵巢。利用Elisa方法检测血清中雌二醇水平,Real time-PCR检测卵巢中激素合成相关基因表达水平。结果:高温促使鸡血清中雌二醇水平显著升高(P<0.05),且卵巢中激素合成相关基因StAR和RUNX2表达量显著下降(P<0.05),当高温情况下饲粮中添加不同水平谷氨酰胺后,35、42日龄鸡血清中雌二醇水平和卵巢中StAR和RUNX2表达量基本恢复到对照组水平,其中42日龄鸡血清中雌二醇水平显著高于对照组(P<0.05),且卵巢中的基因表达水平亦显著高于对照组(P<0.05)。结论:高温环境促使雌鸡生殖性能过快发育,而谷氨酰胺可缓解热应激对雌鸡生殖性能的影响。Objective:To investigate the effect of glutamine(Gln)on reproductive mechanism of chicks under heat stress conditions.Methods:90 healthy 14-day-old female chicks were selected,and randomly divided into 5 groups with 18 chicks in each group,Group I was the control group(room temperature),the chicks were fed with a basic diet.Group II—IV and V were heat stress test groups,with 0%,0.5%,1.0%,1.5%Gln added to the basic diet,respectively.The chicks were pre fed for 7 days,then entered the experimental period that lasted for 21 days.Three chickens were randomly selected at 28,35,42 days of age to collect the blood and ovaries after slaughter.Serum estrogen level was detected by Elisa,and ovarian hormone synthesis-related gene expression was detected by real time-PCR.Results:High temperature significantly increased the serum estrogen level(P<0.05),and decreased the expression of hormone synthesis related genes StAR and RUNX2 in ovary(P<0.05),the levels of estrogens in serum and the expression of StAR and RUNX2 in ovary of 35-day-old and 42-day-old chickens were basically restored to those of the control group when different levels of glutamine were added to the diet at high temperature,however,the level of serum estrogens in 42-day-old chickens was significantly higher than that in the control group(P<0.05),and the gene expression level in ovary was significantly higher than that in the control group(P<0.05).Conclusion:High temperature promotes the rapid development of reproductive performance in female chickens,and glutamine can alleviate the effect of heat stress on reproductive performance.
