淡色库蚊成蚊肠道可培养细菌的分离鉴定与产消化酶活性分析

摘  要:目的为明确淡色库蚊成蚊肠道可培养细菌的组成及产消化酶活性。方法采用传统分离培养的方法,分离纯化淡色库蚊成蚊肠道可培养细菌,通过16S rRNA序列分析进行分子鉴定,利用平板透明圈法筛选产淀粉酶、蛋白酶、纤维素酶和脂肪酶的细菌,通过水解圈(D)与菌落直径(d)的比值,比较不同细菌的产消化酶活性。结果从淡色库蚊成蚊肠道中共分离出11种细菌,其中可产淀粉酶的细菌有6种,产蛋白酶的细菌有7种,4种细菌同时产淀粉酶和蛋白酶,未筛选到产纤维素酶和脂肪酶的细菌。6种细菌的产淀粉酶活性差异具有统计学意义(F=19.451,P<0.0001),其中Pseudochrobactrum的产淀粉酶活性最强;7种细菌的产蛋白酶活性差异有统计学意义(F=5.437,P=0.004),其中金黄杆菌(Chryseobacterium candidae)的产蛋白酶活性最强。结论淡色库蚊成蚊肠道中存在产淀粉酶和蛋白酶的细菌,这些细菌可协助淡色库蚊成蚊消化和分解食物。This study was aimed at determining the composition and digestive enzyme activity of culturable bacteria in the intestinal tract in adult Culex pipiens pallens.Traditional isolation and culture methods were used for the isolation and purification of bacteria from the intestines of adult Cx.pipiens pallens,and 16S rRNA sequencing was performed for identification.The ratio of perhydrolytic circle(D)to colony diameter(d)was used to compare the digestive enzyme activities of the bacteria.A total of 11 bacterial strains were isolated from the intestines of adult Cx.pipiens pallens,among which six were amylase producing bacteria,seven were protease producing bacteria,and four were both amylase producing and protease producing bacteria.No cellulase and lipase producing bacteria were identified.Significant differences in amylase production activity were observed among the six bacteria(F=19.451,P<0.0001),and Pseudochrobactrum had the highest amylase production activity.The protease production activity of the seven bacteria significantly differed(F=5.437,P=0.004),among which Chryseobacterium candidae had the highest protease production activity.Thus,amylase and protease producing bacteria are present in the gut in adult Cx.pipiens pallens,and these bacteria help adult Cx.pipiens pallens digest and decompose food.
