JWH133对右旋糖酐铁过载小鼠脾脏铁代谢影响

摘  要:目的探讨激活大麻素Ⅱ型受体(CB2受体)对右旋糖酐铁处理的铁过载模型小鼠脾脏铁代谢的影响。方法将18只9周龄雄性C57BL/6J小鼠随机分为对照组、右旋糖酐铁组、JWH133(CB2受体激动剂)+右旋糖酐铁组。应用铁检测试剂盒(比色法)检测各组小鼠脾脏铁水平,采用Western blot法检测脾脏转铁蛋白受体1(TFR1)和铁转运蛋白1(FPN1)的表达。结果与对照组相比,右旋糖酐铁组小鼠脾脏总铁含量和Fe 3+含量明显升高(F=12.710、8.352,q=6.980、5.698,P<0.01),Fe 2+含量有增高趋势,但差异无统计学意义;JWH133预处理抑制右旋糖酐铁造成的总铁含量和Fe^(3+)含量的升高(q=4.747、3.687,P<0.05)。与对照组相比,右旋糖酐铁组小鼠脾脏TFR1表达显著升高(F=12.090,q=6.859,P<0.01);JWH133预处理抑制右旋糖酐铁引起的TFR1蛋白表达上调(q=4.419,P<0.05)。3组小鼠脾脏FPN1表达比较差异无显著性(F=1.152,P>0.05)。结论激活CB2受体可以抑制右旋糖酐铁引起的小鼠脾脏铁水平增高以及TFR1蛋白表达上调,CB2受体可能通过调节TFR1蛋白表达介导小鼠脾脏铁的聚集。Objective To investigate the effect of cannabinoid 2 receptor(CB2 receptor)on iron metabolism in the spleen of mice with iron overload treated by iron dextran.Methods A total of 18 male C57BL/6J mice,aged 9 weeks,were randomly divided into control group,iron dextran group,and JWH133(a CB2 receptor agonist)+iron dextran group.An iron assay kit(co-lorimetry)was used to measure the level of iron in the spleen of mice,and Western blot was used to measure the expression of transferrin receptor 1(TFR1)and ferroportin 1(FPN1)in the spleen.Results Compared with the control group,the iron dextran group had significant increases in total iron content and Fe 3+content(F=12.710,8.352,q=6.980,5.698,P<0.01)and a slight increase in Fe 2+content,while JWH133 pretreatment inhibited the increases in total iron content and Fe 3+content caused by iron dextran(q=4.747,3.687,P<0.05).Compared with the control group,the iron dextran group had a significant increase in the expression of TFR1 in the spleen of mice(F=12.090,q=6.859,P<0.01),while JWH133 pretreatment inhibited the upregulation of the expression of TFR1 caused by iron dextran(q=4.419,P<0.05).There was no significant difference in the expression of FPN1 in the spleen of mice between the three groups(F=1.152,P>0.05).Conclusion Activation of CB2 receptors can inhibit the increase in iron level and the upregulation of TFR1 expression in the spleen of mice induced by iron dextran,and CB2 receptor may mediate the accumulation of iron in the spleen of mice by regulating the expression of TFR1 protein.
