芪黄益肾方对糖尿病视网膜病变大鼠视网膜组织病理形态和血-视网膜屏障功能的影响

摘　　要：目的观察芪黄益肾方对糖尿病视网膜病变(DR)大鼠视网膜组织病理形态和血-视网膜屏障功能的影响,探讨其潜在机制。方法采用高脂高糖饮食联合链脲佐菌素腹腔注射法建立DR大鼠模型,将40只成模大鼠随机分为模型组、羟苯磺酸钙组和芪黄益肾方低、中、高剂量组,每组8只,分别灌胃蒸馏水、羟苯磺酸钙混悬液和芪黄益肾方低、中、高浓度溶液,另取8只大鼠作为正常组,灌胃等体积蒸馏水,连续12周。试剂盒检测糖化血红蛋白(HbA1c)含量,HE染色观察视网膜组织病理变化,PAS染色观察视网膜微血管形态,视网膜铺片检测视网膜屏障功能,Western blot检测视网膜组织ZO-1蛋白表达。结果与正常组比较,模型组大鼠HbA1c含量显著增加(P<0.001),视网膜厚度显著减少(P<0.001);视网膜组织细胞排列紊乱,闭塞毛细血管增多,视网膜出现明显渗漏,视网膜组织ZO-1蛋白表达显著降低(P<0.001)。与模型组比较,芪黄益肾方中剂量组大鼠HbA1c含量显著减少(P<0.01),视网膜厚度显著增加(P<0.01);视网膜组织闭塞毛细血管减少,渗漏明显减轻,视网膜组织ZO-1蛋白表达显著升高(P<0.001)。结论芪黄益肾方可降低DR大鼠血糖,减轻视网膜病理损伤,改善视网膜微血管形态,减轻视网膜渗漏,保护血-视网膜屏障功能,其机制可能与上调视网膜组织ZO-1蛋白表达有关。Objective To observe the effects of Qihuang Yishen Prescription on pathological morphology of retina tissue and blood-retina barrier(BRB)function in diabetic retinopathy(DR)rats;To discuss its potential mechanism of action.Methods The DR model was established by using high-fat and high-sugar diet combined with intraperitoneal injection of streptozotocin.Totally 40 DR rats were randomly divided into model group,calcium dobesilate group and Qihuang Yishen Prescription low-,medium-,and high-dosage groups,with 8 in each group.They were administered with distilled water,calcium dobesilate mixture and Qihuang Yishen Prescription low-,midium-,and high-concentration solution respectively for gavage.Another 8 normal rats were gavaged with distilled water as the normal group,for consecutive 12 weeks.The content of HbA1c was detected by kit,the pathological changes of retina tissue were observed by HE staining,the morphology of retinal microvessels was observed by PAS staining,the retinal barrier function was observed by retinal patch,the expression of ZO-1 protein in retinal tissue was detected by Western blot.Results Compared with the normal group,the content of HbA1c in the model group significantly increased(P<0.001),the thickness of retinal tissue significantly decreased(P<0.001);the retinal cells arranged disordered,occluded capillaries increased,and the retina has obviously leakage,the expression of ZO-1 protein significantly decreased(P<0.001).Compared with the model group,the content of HbAlc in Qihuang Yishen Prescription medium-dosage group significantly decreased(P<0.01),the retinal thickness significantly increased(P<0.01);the occluded capillaries in retinal tissue decreased,the leakage was significantly reduced,and the expression of ZO-1 protein significantly increased(P<0.001).Conclusion Qihuang Yishen Prescription can reduce blood sugar in DR model rats,alleviate retinal pathological damage,improve retinal microvascular morphology,reduce retinal leakage,and protect BRB function,which may be achieved by up-re
