摘 要:目的基于IFN-α/JAK1/STAT1信号通路探讨清肺口服液黄酮类组分抗呼吸道合胞病毒(RSV)感染的作用机制。方法36只雌性清洁级BALB/c小鼠随机分为正常组、模型组、利巴韦林组及清肺口服液黄酮类组分低、中、高剂量组(黄酮组分低、中、高剂量组),每组6只。除正常组外,其余组经鼻予RSV病毒液造模,造模成功后给药,各给药组予相应药物灌胃,正常组和模型组予等体积生理盐水,共4 d。HE染色观察小鼠肺组织病理变化,ELISA检测血清α干扰素(IFN-α)含量,RT-PCR检测肺组织RSV-F、Janus酪氨酸蛋白激酶1(JAK1)和信号传导与转录激活因子1(STAT1)mRNA表达,Western blot及免疫组化染色检测肺组织JAK1和STAT1蛋白表达。结果与正常组比较,模型组小鼠肺组织明显充血,有大量炎性细胞浸润,血清IFN-α含量明显增加(P<0.001),肺组织RSV-F、JAK1和STAT1 mRNA表达明显升高(P<0.01),JAK1和STAT1蛋白表达明显升高(P<0.05,P<0.001)。与模型组比较,利巴韦林组及黄酮组分各剂量组小鼠肺组织病理损伤状况明显改善,肺组织RSV-F mRNA表达明显降低(P<0.05);黄酮组分高剂量组血清IFN-α含量明显增加,肺组织JAK1和STAT1 mRNA及蛋白表达明显升高,差异有统计学意义(P<0.05,P<0.01,P<0.001)。结论清肺口服液黄酮类组分具有抗RSV感染作用,其机制可能与上调IFN-α表达,激活JAK1/STAT1信号通路,增强机体免疫功能有关。Objective To explore the mechanism of anti-respiratory syncytial virus(RSV)infection of flavonoids in Qingfei Oral Liquid based on IFN-α/JAK1/STAT1 signaling pathway.Methods Totally 36 female clean-grade BALB/c mice were randomly divided into normal group,model group,ribavirin group and flavonoids in Qingfei Oral Liquid low-,medium-and high-dosage groups,with 6 mice in each group.Except the normal group,the other groups were given RSV via the nose.After successfully modeling,the medication groups were given corresponding therapeutic drugs by gavage,the normal group and the model group were given equal volume of normal saline by gavage,for 4 days.HE staining was used to observe the pathological changes in lung tissue,the content of IFN-αin serum was detected by ELISA,the expression of RSV-F,JAK1 and STAT1 mRNA in lung tissue were detected by RTPCR,Western blot and immunohistochemical staining were used to detect the expression of JAK1 and STAT1 protein in lung tissue.Results Compared with the normal group,the lung tissue of the model group was significantly congested,with a large number of inflammatory cell infiltration,and the content of serum IFN-αincreased significantly(P<0.001),the expression of RSV-F,JAK1 and STAT1 mRNA in lung tissue significantly increased(P<0.01),and the expressions of JAK1 and STAT1 protein significantly increased(P<0.05,P<0.001).Compared with the model group,the pathological damage in lung tissue of the mice in ribavirin group and each dosage of flavonoids in Qingfei Oral Liquid groups significantly improved,the RSV-F mRNA expression in lung tissue significantly decreased(P<0.05);the content of serum IFN-αin flavonoids in Qingfei Oral Liquid high-dosage group increased significantly,the expressions of JAK1 and STAT1 mRNA and protein in lung tissue increased significantly,with statistical significance(P<0.05,P<0.01,P<0.001).Conclusion Flavonoids in Qingfei Oral Liquid can play a role of anti-RSV infection,and its mechanism may be related to the up-regulation of IFN-α,the activation o
