摘 要:目的观察电针对原发性痛经(PDM)大鼠子宫组织ERK/COX-2信号通路相关蛋白及因子表达的影响,探讨电针治疗PDM的可能机制。方法将40只雌性SD大鼠随机分为空白组、模型组、电针组和布洛芬组,每组10只。筛选处于动情间期的大鼠进行造模,除空白组外,其余组连续10 d于股部皮下注射苯甲酸雌二醇,第11日腹腔注射缩宫素诱发疼痛,空白组注射0.9%氯化钠溶液。电针组造模同时电针“关元”“三阴交”,20 min/d,布洛芬组造模同时予布洛芬灌胃,连续10 d。观察各组大鼠行为学变化,HE染色观察子宫组织病理变化,ELISA检测子宫组织前列腺素(PG)F2α和PGE2含量,Western blot检测子宫组织ERK1/2、p-ERK1/2和环氧化酶-2(COX-2)蛋白表达。结果与空白组比较,模型组大鼠扭体次数、扭体评分显著增加(P<0.01);子宫内膜大量空泡样变性、坏死,中性粒细胞浸润,病理评分显著升高(P<0.01);子宫组织PGF2α含量显著增加,PGE2含量显著减少(P<0.01),ERK1/2、p-ERK1/2、COX-2蛋白表达显著升高(P<0.01)。与模型组比较,电针组和布洛芬组大鼠扭体潜伏期显著延长,扭体次数、扭体评分显著降低(P<0.05,P<0.01);子宫内膜较少空泡样变性、坏死,内膜剥脱范围较小,水肿、炎症程度减轻,病理评分显著降低(P<0.05,P<0.01);子宫组织PGF2α含量显著减少,PGE2含量显著增加(P<0.01),ERK1/2、p-ERK1/2、COX-2蛋白表达显著降低(P<0.05,P<0.01)。结论电针可能通过抑制ERK/COX-2信号通路相关蛋白表达调节PG水平,有效缓解PDM大鼠疼痛症状,改善子宫病理损伤。Objective To observe the effects of electroacupuncture on the expression of ERK/COX-2 signaling pathway-related proteins and factors in uterine tissue of rats with primary dysmenorrhea(PDM);To explore the possible mechanisms of electroacupuncture in the treatment of PDM.Methods Totally 40 female SD rats were randomly divided into blank group,model group,electroacupuncture group and ibuprofen group,with 10 rats in each group.The rats in idiestrus were screened for modelling treatment,except for the blank group,all groups were injected subcutaneously with estradiol benzoate in the femur for 10 days,and pain was induced by intraperitoneal injection of indocin on day 11,while the blank group was injected with 0.9%sodium chloride solution.The rats in the electroacupuncture group were simultaneously electroacupunctured with“Guanyuan(CV04)”and“Sanyinjiao(SP06)”for 20 min/d,while the ibuprofen group were simultaneously given ibuprofen by gavage for 10 days.The rats in each group were observed for behavioral changes,HE staining was used to observe pathological changes of uterine tissue,ELISA was used to detect the contents of PG F2αand PGE2 in uterine tissue,the expressions of ERK1/2,p-ERK1/2 and COX-2 protein in uterine tissue were detected by Western blot.Results Compared with the blank group,the rats in model group had significantly increased number of torsions and torsion scores(P<0.01);massive vacuole-like degeneration and necrosis in the endometrium,neutrophil infiltration,significantly higher pathological scores(P<0.01);the content of PGF2αsignificantly increased in uterine tissue and the content of PGE2 significantly decreased(P<0.01),and the expression of ERK1/2,p-ERK1/2 and COX-2 protein significantly increased(P<0.01).Compared with the model group,the rats in electroacupuncture and ibuprofen groups had significantly longer torsion latency,significantly lower number of torsions and torsion scores(P<0.05,P<0.01);less vacuolar-like degeneration and necrosis in the endometrium,less extent of endometrial e
