摘  要:目的基于网络药理学和体内实验探讨黄芪多糖(APS)对三阴性乳腺癌(TNBC)小鼠免疫功能的影响。方法通过网络药理学探索APS治疗TNBC涉及的相关免疫途径,构建荷4T1肿瘤TNBC小鼠模型(分为肿瘤模型组和APS组),给药21 d后,流式细胞术检测小鼠脾脏总细胞中T细胞及其亚群、B细胞、自然杀伤细胞、树突状细胞、髓源性抑制细胞及其亚群、肿瘤相关巨噬细胞的占比和绝对数量,以及其中的免疫细胞活化标记分子CD69在T细胞中、NKp46在自然杀伤细胞中、MHCⅡ在树突状细胞中的表达情况,分析APS对TNBC的免疫调节作用。结果与肿瘤模型组相比,APS组显著增加TNBC小鼠脾脏中的总T细胞,CD8^(+)T细胞、CD4^(+)CD69^(+)T细胞、CD8^(+)CD69^(+)T细胞在脾脏总细胞中的占比;显著增加总T细胞、CD4^(+)T细胞、CD8^(+)T细胞、CD4^(+)CD69^(+)T细胞、CD8^(+)CD69^(+)T细胞的绝对数量。结论APS可通过激活TNBC小鼠的T细胞发挥免疫应答作用。Objective To investigate the effect of Astragalus polysaccharide(APS)on the immune function of triple-negativeb breast cancer(TNBC)tumor-bearing mice based on network pharmacology and in vivo experiments.Methods Network pharmacology was used to explore the possible immune pathways involved in the treatment of TNBC by APS,and 4T1 tumor-bearing TNBC mice were constructed and divided into the model group and APS group.After 21 days of drug administration,the spleens of mice were examined by flow cytometry to analyse the proportion and absolute number of T cells and their subsets,B cells,natural killer cells,dendritic cells,myeloid-derived suppressor cells and their subpopulations,tumor-associated macrophages,as well as the expression of the active markers CD69 in T cell,NKp46 in natural killer cells,and MHCⅡ in dendritic cells in the spleen of tumor-bearing mice 21 days after the administration of APS.Result Compared with the model group,the APS group could significantly increase the total T cells,CD8^(+)T cells,CD4^(+)CD69^(+)T cells,and CD8^(+)CD69^(+)T cells in the spleen of 4T1 tumor-bearing mice,and significantly increased the absolute number of total T cells,CD4^(+)T cells,CD8^(+)T cells,CD4^(+)CD69^(+)T cells and CD8^(+)CD69^(+)T cells.Conclusion APS may exert an immune response by activating T cells in 4T1 tumor-bearing TNBC mice.
