摘  要:多能干细胞因其能够自我更新、自我复制,并具有分化为三胚层来源细胞的潜力而受到生物学家和临床医生的瞩目。上皮细胞是哺乳动物多数器官的组成部分,也是90%的癌症细胞来源,由于体外扩增困难,对其研究深度不如间质细胞。将多能干细胞诱导分化为上皮细胞可以满足人类疾病建模和研究的需求,并有希望治愈临床相关疾病。来自同一胚层的上皮在发育过程中经历相同的发育阶段,参考已有的上皮分化方案,进一步了解不同胚层来源细胞发育历程,为同类型细胞的诱导提供参考。目前人多能干细胞分化为不同胚层来源的人上皮细胞的方法已建立。文章对至今为止的分化方法、鉴定指标及结果进行总结,以供相关人员参考。Biologists and clinicians have been attracted by the pluripotent stem cells because of their ability to self-renew,self-replicate,and potential into triploblast.Most mammalian organs are made up of epithelial cells and 90%of cancer cells are formed by them.Due to the difficulty of in vitro amplification,epithelial cells have not been studied as deeply as stromal cells.Inducing pluripotent stem cells differentiate into epithelial cells would meet the needs of human disease modeling and research and have the promise to cure clinically relevant diseases.The epithelial cells from the same germ layer have experienced the same developmental stage in the process of development.By referring to the existing epithelial differentiation schemes,development process of cells from different germ layers have been understanded and reference for the induction of the same type of cells have been provided.At present,the method of differentiation of human pluripotent stem cells into human epithelial cells of different dermal origin has been established.The differentiation methods,identification indexes and results were summarized by this paper as the reference for relevant personnel.
