摘 要:目的利用网络药理学方法,从整体角度系统分析并预测逍遥散治疗慢性乙型肝炎(CHB)的分子机制,为临床研究及新药研发提供参考。方法检索TCMSP数据库获取逍遥散的活性成分及其相关靶点,并根据口服利用度(OB)和类药性(DL)对活性成分进行筛选。在GeneCards、OMIM数据库中筛选CHB的相关靶点,并与活性成分的靶点取交集,获得逍遥散治疗CHB的潜在作用靶点。在STRING数据库中获得各靶点的相互作用关系,利用Cytoscape 3.8.0软件构建逍遥散-有效活性成分-潜在作用靶点网络及靶点的蛋白互作网络,并进行拓扑结构分析,筛选核心靶点。在R 3.6.2软件中进行潜在作用靶点的基因富集分析。运用BioGPS数据库对靶点进行器官定位。结果通过网络构建及拓扑结构分析发现,逍遥散主要通过113个有效活性成分干预155个潜在靶点来治疗CHB,有效活性成分中以槲皮素、木犀草素等黄酮类成分为主,核心靶点为ESR1、CDK2、RXRA、RELA、AKT1、MAPK14、MAPK1等。基因富集分析显示逍遥散主要通过干预氧化应激相关生物学过程及调控PI3K-Akt、MAPK等信号通路,发挥治疗CHB的作用。器官定位结果表明逍遥散治疗CHB主要作用于肝系与脾系。结论逍遥散主要通过下调HBV的转录与复制、调节免疫反应及炎症反应、调控细胞周期等发挥治疗CHB的作用。Objective The mechanism of Xiaoyao Powder in the treatment of chronic hepatitis B(CHB)was systematically predicted and analyzed by network pharmacology to provide a reference for clinical research and the development of new drugs.Methods The TCMSP database was searched to obtain the active ingredients of Xiaoyao Powder and their related targets,and the active ingredients were screened according to oral bioavailability(OB)and drug-like properties(DL).The relevant targets of CHB were screened in GeneCards,OMIM database,and intersected with the targets of active ingredients to obtain the potential targets of action of Xiaoyao Powder for CHB.The interaction relationship of targets was obtained in STRING database,and Cytoscape 3.8.0 software was used to construct the network of Xiaoyao Powder-active ingredients-potential action targets and protein interaction network of targets,and topology analysis was performed to screen the core targets.Gene enrichment analysis of potential targets was performed in R 3.6.2 software.Organ localization of targets was performed using the BioGPS database.Results The network construction and topological analysis revealed that Xiaoyao Powder mainly interfered with 155 potential targets through 113 active ingredients for the treatment of CHB,and the active ingredients were mainly flavonoids such as quercetin and lignan,and the core targets were ESR1,CDK2,RXRA,RELA,AKT1,MAPK14,MAPK1,etc.The gene enrichment analysis showed that Xiaoyao Powder mainly interfered with oxidative stress-related biological processes and regulated PI3K-Akt,MAPK and other signaling pathways to treat CHB.The organ localization results showed that the treatment of CHB with Xiaoyao Powder mainly acted on the hepatic and splenic tracts.Conclusion The treatment of CHB with Xiaoyao Powder mainly involves down-regulation of HBV transcription and replication,regulation of immune response and inflammatory response,and regulation of cell cycle.
