摘 要:YUCCA基因编码生长素合成相关酶,在植物的生长发育过程中发挥重要作用。利用Genevestigator软件分析获得玉米YUCCA(ZmYUC)基因的转录表达谱,结果显示,ZmYUC1、ZmYUC2、ZmYUC4、ZmYUC5、ZmYUC10和ZmYUC12等多个ZmYUC基因在特定组织中高水平表达,ZmYUC7和ZmSPI1等基因在所检测的组织中几乎均有表达,暗示ZmYUC基因在玉米不同生长发育过程均发挥重要作用。通过PlantCare软件分析ZmYUC基因启动子区的顺式作用元件,获得其在逆境胁迫方面的顺式作用元件,利用实时定量PCR方法对ZmYUC基因在冷(4℃)、热(40℃)、盐(250 mmol/L的NaCl)和干旱(15%的PEG6000)处理下的表达量变化进行检测,结果显示,ZmYUC5和ZmYUC8在冷胁迫处理下表达量显著增加,可能参与了植物的冷驯化;热胁迫处理后,大多数ZmYUC基因的表达模式发生了明显变化,其中,ZmYUC8对热信号反应迅速,可能在抵抗热胁迫中发挥关键作用,ZmYUC11热胁迫处理后其表达下调。多数ZmYUC基因在干旱胁迫处理后表达模式未见明显变化;盐胁迫处理后,只有ZmYUC3、ZmYUC4、ZmYUC5、ZmYUC6和ZmYUC8等基因的表达有所变化,其他基因表达无明显变化。YUCCA genes,encoding enzymes associated with auxin biosynthesis,play essential roles in growth and development of plants by contributing to the local pool of free auxins.The transcriptional profile of maize YUCCA(ZmYUC)gene was analyzed by the Genevestigator software.The results showed that several ZmYUC genes including ZmYUC1,ZmYUC2,ZmYUC4,ZmYUC5,ZmYUC10 and ZmYUC12 were expressed in high level in specific tissues,ZmYUC7 and ZmSPI1 genes were almost all expressed in the tissues tested,which suggesting that ZmYUC genes play important roles in different growth and development processes of maize.The cis-acting elements of ZmYUC promoter region were analyzed by PlantCare software to obtain cis-acting elements in stress.Then,realtime quantitative PCR was used to detect the expression changes of ZmYUC gene under cold(4℃),heat(40℃),salt(250 mmol/L NaCl)and drought(15%PEG6000),and analyze the expression patterns of ZmYUC gene under different tissues and organs and different stress treatments.The results showed that the expression levels of ZmYUC5 and ZmYUC8 significantly increased under cold stress,which may be involved in cold acclimation of plants.After heat stress treatment,the expression patterns of most ZmYUC genes changed significantly.ZmYUC8 was rapidly responsive to heat signal and may play a key role in resistance to heat stress,while ZmYUC11 was down-regulated after heat stress treatment.The expression pattern of most ZmYUC genes did not change significantly after drought stress treatment after salt stress treatment,only the expressions of ZmYUC3,ZmYUC4,ZmYUC5,ZmYUC6 and ZmYUC8 genes were changed,while the expressions of other genes were not significantly changed.
