两种固定液对鸭小肠肥大细胞染色效果的比较

摘　　要：为了探讨不同固定液对鸭小肠肥大细胞甲苯胺蓝染色效果的影响及鸭小肠内肥大细胞的数量和分布情况,试验选择5只100日龄的健康淮南麻鸭公鸭为研究对象,取鸭只十二指肠、空肠和回肠组织样品,均分为两份,分别置于10%中性福尔马林固定液和卡恩氏(Carnoy)固定液中固定,常规制作石蜡切片,切片经甲苯胺蓝染色后,观察小肠肥大细胞染色效果和肥大细胞形态、数量及分布,并进行统计分析。结果表明:Carnoy固定液对肥大细胞的染色效果优于10%中性福尔马林固定液,染色评分极显著高于10%中性福尔马林固定液(P<0.01);Carnoy固定液固定的十二指肠、空肠、回肠肥大细胞数量均极显著高于10%中性福尔马林固定液(P<0.01)。鸭十二指肠、空肠和回肠中肥大细胞形态和数目不同,在黏膜固有层血管、腺管周围的结缔组织中分布较多;空肠中肥大细胞数量最多,形态多样;十二指肠和回肠中肥大细胞数量递减,形态多为圆形。说明与10%中性福尔马林固定液相比,经Carnoy固定液固定的鸭小肠组织染色后,可清晰显示肥大细胞的形态分布。To investigate the effects of different fixatives on the toluidine blue staining of small intestinal mast cells in ducks and the distribution of small intestinal mast cells in ducks,five 100-day-old healthy male Huainan ducks were selected as the study subjects.The duodenum,jejunum and ileum tissue samples of ducks were collected and divided into two parts,which were fixed in 10%buffered formalin and Carnoy's fluid fixatives,respectively.Then,paraffin sections were routinely prepared.The sections were stained with toluidine blue,the morphology,number and distribution of mast cells in each tissue were observed under light microscopy,and statistical analysis was performed.The results showed that the staining effects of Carnoy's fixative on mast cells was better than that of 10%neutral formalin fixative,and the staining evaluation score was significantly higher than that of 10%formalin solution fixative(P<0.01).The number of mast cells in the duodenum,jejunum and ileum fixed with Carnoy's fixative solution was significantly higher than that with 10%neutral formalin fixative solution(P<0.01).The morphology and number of mast cells in duck duodenum,jejunum and ileum were different,and they were more distributed in the connective tissue around the blood vessels and glandular ducts in the lamina propria of mucous membrane.The number of mast cells in the jejunum of duck was the largest and varied,while the number of mast cells in the duodenum and ileum decreased,and most of them were round.The results indicated that the morphologic distribution of mast cells could be clearly displayed after staining of small intestine tissues fixed with Carnoy's fixative compared with 10%formalin solution fixative.
