摘  要:目的:探讨蛇床子素对结肠癌细胞增殖、迁移和侵袭的影响,以及对Toll样受体4(TLR4)/髓样细胞分化因子88(MyD88)/核转录因子κB(NF-κB)信号通路的调节作用。方法:第3代对数生长期结肠癌SW480细胞用含不同浓度(0、50、100、200μmol/L)蛇床子素的培养基培养,采用MTT法检测细胞存活率,平板克隆实验检测细胞增殖情况,Transwell实验检测细胞迁移和侵袭能力,蛋白印迹法(Western blotting)检测TLR4、MyD88和NF-κB蛋白相对表达量。结果:50、100、200μmol/L蛇床子素组细胞存活率、TLR4蛋白相对表达量、MyD88蛋白相对表达量、NF-κB蛋白相对表达量、细胞菌落数、迁移细胞数和侵袭细胞数均低于0μmol/L蛇床子素组(P<0.05);100、200μmol/L蛇床子素组细胞存活率、TLR4蛋白相对表达量、MyD88蛋白相对表达量、NF-κB蛋白相对表达量、细胞菌落数、迁移细胞数和侵袭细胞数均低于50μmol/L蛇床子素组(P<0.05);200μmol/L蛇床子素组细胞存活率、TLR4蛋白相对表达量、MyD88蛋白相对表达量、NF-κB蛋白相对表达量、细胞菌落数、迁移细胞数和侵袭细胞数均低于100μmol/L蛇床子素组(P<0.05)。结论:蛇床子素可抑制结肠癌SW480细胞增殖、迁移和侵袭,其机制可能是抑制TLR4/MyD88/NF-κB信号通路。Objective:To investigate the effects of osthole on proliferation,migration and invasion of colon cancer cells,and the regulation of Toll-like receptor 4(TLR4)/myeloid cell differentiation factor(MyD88)/nuclear transcription factorκB(NF-κB)signaling pathway.Methods:The third passage of SW480 cells in logarithmic growth phase were cultured in medium containing different concentrations of osthole(0,50,100,200μmol/L).MTT assay was used to detect cell viability,plate clone assay was used to detect cell proliferation.Transwell assay was used to detect cell migration and invasion.The relative expression levels of TLR4,MyD88 and NF-κB protein were detected by western blotting.Results:The cell survival rate,the relative expression of TLR4,MyD88 and NF-κB protein,the number of cell colony,migrating cells and invading cells in the 50,100 and 200μmol/L osthole groups were lower than those in 0μmol/L osthole group(P<0.05).The cell survival rate,the relative expression of TLR4,MyD88 and NF-κB protein,the number of cell colony,migrating cells and invading cells in the 100 and 200μmol/L osthole groups were lower than those in 50μmol/L osthole group(P<0.05).The cell survival rate,the relative expression of TLR4,MyD88 and NF-κB protein,the number of cell colony,migrating cells and invading cells in the 200μmol/L osthole group were lower than those in 100μmol/L osthole group(P<0.05).Conclusion:Osthole can inhibit the proliferation,migration and invasion of colon cancer SW480 cells,possibly through inhibiting TLR4/MyD88/NF-κB signaling pathway.
