茵陈四苓汤对酒精性肝损伤大鼠肠黏膜屏障ZO-1及occludin的影响

摘　　要：目的:观察茵陈四苓汤对酒精性肝损伤大鼠肠黏膜屏障血清闭锁小带蛋白-1(ZO-1)及闭锁蛋白(occludin)影响。方法:用高脂饮食加白酒灌胃建立酒精性肝损伤大鼠模型,将造模成功的SD大鼠随机分为模型组、双环醇组及茵陈四苓汤组,并选取同批次不造模大鼠为空白组。药物干预4周后,观察各组大鼠肝功能(AST、ALT)、血脂(TC、TG)、血浆内毒素水平、肠黏膜屏障ZO-1、occludin表达及肝脏病理形态学的改变。结果:与模型组比较,茵陈四苓汤组大鼠ALT、AST、TC、TG、LPS水平均降低(P<0.05或P<0.01);与模型组比较,茵陈四苓汤组大鼠ZO-1、occludin表达升高(P<0.01);与模型组比较,茵陈四苓汤组大鼠肝脏组织及肝细胞脂肪变性均有改善,程度不等。结论:茵陈四苓汤对酒精性肝损伤大鼠的肝功能、血脂具有一定的调节作用,其作用机制与增加肠黏膜ZO-1、occludin表达、保护肠黏膜屏障、降低内毒素水平有关。Objective:To investigate the effect of YinChen Siling Decoction on ZO-1 and occludin in intestinal barrier in rats with alcoholic liver injury.Methods:A rat model of alcoholic liver injury was established by gavage with high-fat diet and white wine,and these SD rats with successful modeling were divided into model group,bicyclol group and YinChen Siling Decoction group,and the same batch of rats without modeling were selected as normal group.After 4 weeks of drug intervention,the rats in each group were observed for changes in liver function(AST,ALT),blood lipids(TC,TG),plasma endotoxin levels,intestinal mucosal barrier ZO-1,occludin expression and liver pathomorphology.Results:Compared with the model group,the levels of ALT,AST,TC,TG and LPS were reduced in the YinChen Siling Decoction group(P<0.05 or P<0.01).Compared with the model group,the expression of ZO-1 and occludin increased in the YinChen Siling Decoction group(P<0.01).Compared with the model group,the liver tissue and hepatocyte steatosis were improved in the YinChen Siling Decoction group to different degrees.Conclusion:YinChen Siling Decoction has certain regulatory effects on liver function and blood lipids in rats with alcoholic liver injury,which is related to increasing the expression of ZO-1 and occludin in the intestinal mucosa,protecting the intestinal mucosal barrier and reducing the level of endotoxin.
