摘 要:目的探讨左归丸对睡眠剥夺大鼠的认知功能的作用及相关分子机制的研究。方法选择从2021年4月开始,进行为期9周的试验。将40只远交群(SD)大鼠随机分为对照组、模型组(睡眠剥夺组)、左归丸低、中、高剂量组,构建大鼠Morris水迷宫实验模型。通过水迷宫检测大鼠空间学习记忆能力;试剂盒法检测大鼠海马组织中丙二醛(MDA),谷胱甘肽过氧化物酶(GSH-Px)和超氧化物歧化酶(SOD)表达水平,以及血清中炎性因子白介素-1β(IL-1β)、白介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)含量;免疫印迹(Western blot)法检测海马组织中IL-1β、IL-6、TNF-α和核因子-κB(NF-κB)的蛋白表达水平。结果与对照组比较,模型组大鼠游泳总路程和逃避潜伏期明显延长(P<0.05);原象限停留时间和平台穿越次数明显减少(P<0.05);血清中IL-1β、IL-6和TNF-α含量显著升高(P<0.05);MDA含量明显增加(P<0.05),而GSH-Px和SOD含量明显降低(P<0.05);海马组织中IL-1β、IL-6、TNF-α和NF-κB相对蛋白表达水平显著上调(P<0.05)。与模型组比较较,左归丸干预后大鼠游泳总路程和逃避潜伏期显著减少,原象限停留时间和平台穿越次数显著增加(P<0.05);血清中IL-1β、IL-6和TNF-α含量显著下降,MDA含量显著降低,而GSH-Px和SOD含量显著增加(P<0.05);IL-1β、IL-6、TNF-α以及NF-κB蛋白表达水平均有不同程度的下调(P<0.05)。结论左归丸能够改善睡眠剥夺引起的大鼠认知功能障碍,其机制可能与降低炎性反应、提高抗氧化水平有关。Objective To explore the effect of Zuogui Pill on cognitive function in sleep-deprived rats and related research of its molecular mechanism.Methods This 9-week study was conducted from April,2021.Sprague Dawley(SD)rats(n=40)were randomly divided into control group,model group(sleep-deprived group),Zuogui Pill low-,medium-and high-dose groups.A Morris water maze model was established.Water maze experiment was carried out to evaluate the spatial learning and memory ability of rats.The reagent kit method was performed to detect the levels of malondialdehyde(MDA),Glutathione peroxidase(GSH-Px)and supper oxide dismutase(SOD)in hippocampus of rats,as well as inflammatory cytokines including interleukin-1beta(IL-1β),interleukin 6(IL-6)and tumor necrosis factor alpha(TNF-α).Western blot analysis was conducted to analyze the protein expression levels of IL-1β,IL-6,TNF-αand NF-kappa B(NF-κB)in hippocampus.Results Compared with the control group,the model group had significant prolonged total swimming distance and escape latency(P<0.05),significant reduced original quadrant residence time and platform crossing times(P<0.05),significant elevated serum levels of IL-1β,IL-6,TNF-α(P<0.05),significant increased the content of MDA(P<0.05),significant decreased the content of GSH-Px and SOD(P<0.05),significantly up-regulated relative protein expression levels of IL-1β,IL-6,TNF-αand NF-κB in hippocampus(P<0.05).Compared with the model group,Zuogui Pill groups had significant shortened total swimming distance and escape latency(P<0.05),significant increased original quadrant residence time and platform crossing times(P<0.05),significantly decreased serum levels of IL-1β,IL-6,TNF-α(P<0.05),significantly decreased the content of MDA(P<0.05),significantly increased the content of GSH-Px and SOD(P<0.05),and significantly down-regulated relative protein expression levels of IL-1β,IL-6,TNF-αand NF-κB in hippocampus(P<0.05).Conclusion Zuogui Pill can improve cognitive dysfunction induced by sleep deprivation,which may be rel
