摘  要:【目的】分析青花菜BoWHY2基因的序列特征和分子进化概况,检测高温胁迫下的表达特征,为揭示青花菜BoWHY2基因高温胁迫下的应答机制提供依据。【方法】利用PCR技术克隆青花菜BoWHY2基因序列,并分析其序列特征、分子进化和高温胁迫下的表达特征。【结果】克隆获得青花菜BoWHY2基因,其CDS区域长度为723 bp,推导编码240个氨基酸。该蛋白分子量为26.2 kD,属于碱性蛋白,疏水性较好,理论等电点为9.48,其表面有1个中心空穴并形成带电的内腔。青花菜BoWHY2基因在高温胁迫下的相对表达量处理3 h时下降,处理12 h时升高。青花菜BoWHY2基因与甘蓝和油菜的进化关系较近。【结论】青花菜BoWHY2基因与甘蓝和油菜的进化关系较近,保守性较强;BoWHY2基因在高温胁迫下其表达量持续升高,对高温胁迫能够做出正向反应。【Objective】The sequence feature and molecular evolution of broccoli BoWHY2 gene are analyzed and its expression characteristics is detected under high temperature stress to reveal the response mechanism of broccoli BoWHY2 gene to high temperature stress.【Method】The sequence feature and molecular evolution of broccoli BoWHY2 gene and its expression characteristics under high temperature stress are analysed by cloning the sequence of broccoli BoWHY2 gene through PCR technology.【Result】The CDS length,deduced coding,protein molecular weight(alkaline protein with good hydrophobicity)and theoretical isoelectric point of the cloned broccoli BoWHY2 gene are 723 bp,240 amino acids,26.2 kD and 9.48 respectively.There is a central hole with an electriferous interspace on the protein surface.The relative expression level of broccoli BoWHY2 gene decreases after 3 h high temperature stress and increases after 12 h high temperature stress.The broccoli BoWHY2 gene has the closer evolution relationship with Brassica oleracea and Brassica napus.【Conclusion】There is a closer evolution relationship between broccoli BoWHY2 gene with a strong conservatism and B.oleracea,B.napus.The expression level of broccoli BoWHY2 gene rises continuously under high temperature stress,which means that broccoli BoWHY2 gene can react the forward reaction to high temperature stress.
