摘 要:目的研究卵巢早衰小鼠血清Merlin蛋白水平变化及其与血清E2、FSH、LH的关系。方法120只6周龄KM雌性小鼠按随机原则分为三组即实验A组,实验B组和空白对照组。实验A组小鼠经腹腔注射环磷酰胺120 mg/kg,观察10 d,建POF模型;实验B组小鼠经灌胃给予雷公藤多甙片(Tripterygium wilfordii polyglycoside)0.6 mg/100g,1次/d,连续10 d,构建POF模型;空白对照组等剂量生理盐水灌胃,1次/d,共10 d。采血测血清E2、FSH、LH和Merlin蛋白浓度,比较各组建模前后及三组间建模后Merlin蛋白表达水平;同时分析建模后实验组血清Merlin蛋白水平与血清E2、FSH、LH水平的相关性。结果建模后三组间血清Merlin蛋白水平有明显差异(F=80.41,P<0.01),经组间两两比较,实验A组和实验B组小鼠血清的Merlin蛋白水平[(3.76±1.15)ng/mL;(3.85±1.31)ng/mL]与对照组有明显下降(t=11.18&10.78),差异有统计学意义(P<0.01)。实验A组和实验B组建模后Merlin蛋白水平分别较建模前差异均有统计学意义(t=12.53&11.14;P<0.01)。Pearson相关分析显示,建模后实验A组和实验B组血清Merlin蛋白水平与各自E2水平呈负相关(r=-0.812&-0.789),差异有统计学意义(P<0.01);与各自FSH和LH水平呈正相关(r=0.715&0.722;0.732&0.741),差异均具有统计学意义(P<0.01)。结论Merlin蛋白可能参与了卵巢早衰的激素调控,且对卵泡生长发育产生了负性调控作用。Objective To study the serum level of Merlin protein in mice with premature ovarian failure and its relationships with the levels of E2,FSH,and LH.Methods One hundred twenty 6-week-old female KM mice were randomly divided into experimental group A experimental group B and blank group.The mice in experimental group A were treated with intraperitoneal injection of cyclophosphamide(120 mg/kg)and observed for 10 d;the mice in experimental group B were treated with pouring Tripterygium wilfordii polyglycoside tablets(TWP)(0.6 mg/100 g)into stomach for 10d while the rats in blank group were treated with pouring normal saline of the same volume into stomach for 10 d.Before and after modeling,the serum was collected to measure E2,FSH,LH,and Merlin protein concentrations.The Merlin protein expressions in each group were compared after modeling.The correlation between Merlin protein and E2,FSH,LH were also carried out after modeling.Results There were significant differences in the serum Merlin protein levels between the three groups after modeling(F=80.41,P<0.01);Performing pairwise comparisons the serum Merlin protein levels of mice in experimental group A(3.76±1.15 ng/mL)and experimental group B(3.85±1.31 ng/mL)were decreased compared with the control group(t=11.18&10.78;P<0.01).The levels of Merlin protein in experimental group A and experimental group B were significantly different than those before modeling respectively after modeling(t=12.53&11.14;P<0.01).Pearson correlation analysis showed that the serum Merlin protein levels of experimental group A and experimental group B were negatively correlated with E2 levels(r=-0.812&-0.789;P<0.01)and positively correlated with FSH and LH levels(r=0.715&0.722;0.732&0.741;P<0.01)after modeling.Conclusion Merlin protein might be involved in the hormonal regulation of premature ovarian failure and has a negative regulatory effect on the growth and development of follicles.
