摘 要:目的:应用生物信息学分析软件预测并分析人多聚腺苷酸结合蛋白4(PABPC4)和人内质网二硫化物氧化酶1α(ERO1α)蛋白结构和功能,并结合该课题组前期实验结果分析其与子痫前期的关系,为预防子痫前期不良妊娠结局提供新的诊疗思路。方法:利用NCBI数据库、Protparam软件、DeepTMHMM、SignalP-5.0、Cell-Ploc等生物信息学预测软件对PABPC4和ERO1α蛋白理化性质、信号肽、跨膜区、亚细胞定位分析,选取String及IEDB等在线网站分析PABPC4和ERO1α的相互作用蛋白及蛋白优势表位预测。结果:PABPC4蛋白共编码660个氨基酸的,相对分子质量为72390.64道尔顿,为不稳定的亲水性蛋白,无跨膜区,无信号肽,亚细胞定位在细胞质、细胞核上,有多种蛋白与之相互作用。ERO1α蛋白共编码468个氨基酸的,相对分子质量为54392.66道尔顿,为不稳定的亲水性蛋白,无跨膜区,有信号肽,亚细胞定位在内质网上,有多种蛋白与之相互作用。结论:生物信息学预测分析PABPC4和ERO1α含有多个磷酸化位点和B细胞优势表位,并且结合前期与子痫前期(preeclampsia,PE)的研究,为进一步针对PABPC4和ERO1α研究PE设计多肽提供新靶点。Objective To predict and analyze the structure and function of human polyadenylate-binding protein 4(PABPC4)and human endoplasmic reticulum disulfide oxidase 1a(ERO1a)protein by using bioinformatics software,and to analyze their relationship with preeclampsia in conjunction with the results of previous experiments of the group,in order to provide new diagnostic ideas for the prevention of adverse pregnancy outcomes in preeclampsia.Methods Bioinformatics prediction software such as NCBI database,Protparam software,DeepTMHMM,SignalP-5.0 and Cell-Ploc were used to analyze the physicochemical properties,signal peptide,transmembrane region and subcellular localization of PABPC4 and ERO1a proteins,and selected online sites such as String and IEDB to analyze PABPC4 and ERO1a interacting proteins and protein dominant epitope prediction.Results PABPC4 protein encodes 660 amino acids with a relative molecular mass of 72,390.64 daltons,is an unstable hydrophilic protein with no transmembrane region and no signaling skin,and is subcellularly localized to the cytoplasm and nucleus.ERO1a protein encodes 468 amino acids with a relative molecular mass of 54,392.66 daltons,is an unstable hydrophilic protein with no transmembrane region,has a signaling skin,is subcellularly localized to the endoplasmic reticulum,and has multiple protein interactions with.Conclusion Bioinformatic prediction analysis of PABPC4 and ERO1 contains multiple phosphorylation sites and B-cell dominant epitopes,and in combination with preeclampsia(PE)studies,provides new targets for further PE design multi-skin studies for PABPC4 and EROla.
