脑缺血大鼠海马区和皮质区转铁蛋白受体的表达及脑泰方干预作用研究

摘  要:目的 研究脑缺血大鼠转铁蛋白受体(TFR)的表达及中药脑泰方干预作用,阐明脑缺血后铁代谢失调的机制及脑泰方通过调节细胞内铁代谢,保护神经元。方法 随机将SD大鼠分为假手术组、模型组,制备大鼠脑缺血模型,通过免疫荧光检测海马区和皮质区TFR表达情况。随机将SD大鼠分为低、中、高剂量组,通过蛋白质印迹法(WB)检测各剂量组海马区和皮质区TFR蛋白表达情况,通过聚合酶链式反应(PCR)检测m RNA表达情况。结果 大鼠大脑中动脉栓塞术后免疫荧光TFR表达:与假手术组相比,模型组表达明显增高(P <0.05)。TFR WB检测显示:与模型组比较,脑泰方低、中、高剂量组免疫荧光评分均降低(P <0.05),其中,高剂量组表达明显降低(P <0.05)。TFR m RNA表达:与模型组比较,脑泰方低、中、高剂量组m RNA评分均降低(P <0.05),其中,中、高剂量组表达明显降低(P <0.05)。结论 铁调节失衡可能是脑缺血海马和皮质神经元损伤发生的机制之一,同时,脑缺血后益气活血中药脑泰方能通过干预海马和皮质的TFR的表达,起到维持脑铁平衡、保护脑神经的作用。Objective To study the expression of murine transferrin receptor TFR and the intervention effect of traditional Chinese medicine Naotai formula,to elucidate the mechanism of iron metabolism disorder after cerebral ischemia and the protection of neurons by regulating intracellular iron metabolism.Methods SD rats were randomly divided into sham surgery group and model group,rat cerebral ischemia models were prepared,and TFR expression in hippocampal and cortical regions was detected by immunofluorescence.SD rats were randomly divided into low,medium,and high dose groups,and the expression of TFR protein in the hippocampal and cortical regions of each dose group was detected by Western blotting,and the expression of mRNA was detected by PCR.Results The expression of immunofluorescent TFR in rats after middle cerebral artery embolization was significantly increased in the model group compared with the sham group(P<0.05).TFR WB detection showed that compared with the model group,the immunofluorescence score of the low,medium and high dose groups of cerebral Naotai formula decreased(P<0.05),and the expression of the high-dose group was significantly reduced(P<0.05).TFR mRNA expression:Compared with the model group,the mRNA score of the low,medium,and high dose groups was reduced(P<0.05),and the expression in the medium-high dose group was significantly reduced(P<0.05).Conclusion Iron regulation imbalance may be one of the mechanisms of cerebral ischemic hippocampal and cortical neuronal damage,and at the same time,Chinese medicine Naotai formula can maintain cerebral iron balance and protect cranial nerves by intervening in the expression of TFR of hippocampus and cortex.
