摘  要:目的探究甲巯咪唑联合双膦酸盐对甲亢性骨质疏松大鼠骨密度的影响。方法选取60只SPF级雄性SD大鼠,随机分为5组:对照组(生理盐水灌胃)、模型组(左甲状腺素灌胃)、甲巯咪唑组(左甲状腺素灌胃,甲巯咪唑治疗)、双膦酸盐组(左甲状腺素灌胃,双膦酸盐治疗)和联合组(左甲状腺素灌胃,甲巯咪唑联合双膦酸盐治疗),每组12只。检测各组大鼠血清中甲状腺功能指标游离三碘甲状腺原氨酸(FT3)、游离甲状腺素(FT4)含量,骨代谢指标钙(Ca)、抗酒石酸盐酸性磷酸酶(TRAP)、碱性磷酸酶(ALP)、骨钙素(BGP)含量。检测各组大鼠骨矿物质含量、骨密度以及股骨骨折应力和压碎力。结果与对照组比较,其余各组大鼠FT3、FT4含量均显著升高(F=18.560、35.120,P均<0.05),Ca含量显著降低(F=4.607,P<0.05),TRAP、ALP、BGP含量均显著升高(F=22.850、15.790、18.480,P均<0.05),骨矿物质含量、骨密度、股骨骨折应力和压碎力显著降低(F=8.565、2.567、13.790、21.490,P均<0.05),差异均有统计学意义。与模型组比较,甲巯咪唑组、双膦酸盐组和联合组大鼠FT3、FT4、TRAP、ALP、BGP含量均显著降低,骨矿物质含量、骨密度、股骨骨折应力和压碎力均显著升高,差异均有统计学意义(P均<0.05),但对钙含量无明显反应(P>0.05)。甲巯咪唑治疗可显著降低FT3、FT4、TRAP、ALP、BGP含量,提高骨矿物质含量、骨密度、股骨骨折应力和压碎力。双膦酸盐治疗可显著降低TRAP、ALP、BGP含量,提高骨矿物质含量、骨密度、股骨骨折应力和压碎力。甲巯咪唑和双膦酸盐联合治疗可进一步改善以上各指标。结论甲巯咪唑联合双膦酸盐治疗甲亢性骨质疏松大鼠,可显著改善大鼠甲状腺功能,并显著改善大鼠骨质疏松情况。联合用药疗效显著优于单独用药。Objective To investigate the effect of methimazole combined with bisphosphonates on bone mineral density in rats with hyperthyroid osteoporosis.Methods Sixty SPF male SD rats were randomly divided into five groups with twelve rats in each group.The groups were as follows:control group(normal saline gavage),model group(left thyroxine gavage),methimazole group(left thyroxine gavage,methimazole treatment),bisphosphonate group(left thyroxine gavage,bisphosphonate treatment)and combination group(left thyroxine gavage,methimazole combined with bisphosphonate treatment).The contents of thyroid function indexes free triiodothyronine(FT3)and free thyroxine(FT4)and bone metabolism indexes calcium(Ca),tarrtrate-resistant acid phosphatase(TRAP),alkaline phosphatase(ALP),bone glaprotein(BGP)in serum were measured.The bone mineral content,bone mineral density,femoral fracture stress and crushing force were measured.Results Compared with the control group,the contents of FT3 and FT4 increased significantly(F=18.560,35.120,all P<0.05);the content of Ca decreased significantly(F=4.607,P<0.05);the contents of TRAP,ALP and BGP increased significantly(F=22.850,15.790,18.480,all P<0.05);the content of bone minerals and bone mineral density decreased significantly,and the stress and crushing force of femoral fracture decreased significantly(F=8.565,2.567,13.790,21.490,all P<0.05).Compared with the model group,the contents of FT3,FT4,trap,ALP and BGP in methimazole group,bisphosphonate group and combined group decreased significantly,the content of bone minerals and bone mineral density increased significantly,and the stress and crushing force of femoral fracture increased significantly(P<0.05).There was no significant changes in Ca content(P>0.05).Methimazole treatment could significantly reduce the contents of FT3,FT4,TRAP,ALP and BGP,increase the content of bone minerals,bone mineral density,femoral fracture stress and crushing force.Bisphosphonate treatment could significantly reduce the contents of trap,ALP and BGP,increase bone m
