摘 要:以红托竹荪菌柄为研究对象,采用响应面法优化红托竹荪菌柄多糖的提取工艺,通过Sevage法及透析法去除蛋白和小分子,获得一种具有一定抗氧化活性的红托竹荪菌柄多糖(Dictyophora rubrovalvata stipe polysaccharide,Drsp)。利用傅里叶红外光谱(Fourier transform infrared spectrometer,FTIR)和离子色谱对Drsp进行结构表征,并进一步通过体外和细胞内抗氧化试验检测Drsp的抗氧化活性。结果表明,当料液比为1:25(g/mL)、提取时间为3 h、提取温度为80℃、提取次数为2次时,多糖的提取率相对较高,在此条件下菌柄多糖的平均得率为(9.68±0.08)%;FTIR结果表明,Drsp是一种含β-糖苷键的吡喃型多糖;离子色谱测定其单糖组成为岩藻糖(1.65%)、半乳糖(6.98%)、葡萄糖(80.32%)、木糖(0.38%)、甘露糖(10.67%);体外抗氧化试验表明,Drsp具有较好的抗氧化性(ABTS:IC_(50)=(325.15±5.5)μg/mL;FRAP:IC_(50)=(613.1±39.1)μg/mL);细胞抗氧化试验表明,Drsp降低了H_(2)O_(2)诱导的HaCaT细胞中LDH的分泌和MDA含量,并提高了SOD活力,从而减轻了细胞氧化应激损伤。该研究为红托竹荪菌柄多糖的综合性开发与应用提供科学数据。In this study,the extraction process of polysaccharide from stipe of Dictyophora rubrovalvata was optimized by response surface method.The proteins and small molecules were removed by Sevage and dialysis method to obtain D.rubrovalvata stipe polysaccharide(Drsp)with certain antioxidant activity.The structure of Drsp was characterized by Fourier transform infrared spectroscopy and ion chromatography,and its antioxidant activity was evaluated by in vitro and intracellular antioxidant assays.The findings indicated that when the solid to liquid ratio was 1:25,the extraction period was 3 h,the extraction temperature was 8o ℃,and the extraction times were twice,the extraction rate of polysaccharide was reasonably high.The average polysaccharide output in these circumstances was(9.68±0.08)%.Drsp is a pyran-type polysaccharide that contains β-glycosidic bonds,as revealed by its Fourier transform infrared spectrum;ion chromatography revealed its monosaccharide composition to be fucose(1.65%),galactose(6.98%),glucose(80.32%),xylose(0.38%)and mannose(10.67%).In vitro tests,Drsp showed significant antioxidant activity(ABTS:IC_(50)=(325.15±5.5)g/mL;FRAP:IC_(50)=(613.1±39.1)g/mL).In the cell assays,Drsp reduced LDH secretion and MDA content in H_(0)O_(2)-induced HaCaT cells while increasing SOD activity,reducing oxidative stress damage in cells.The scientific date given by this study has led to substantial development and application of the stipe polysaccharide of D.rubrovalvata.
