摘 要:目的 利用生物信息学筛选急性ST段抬高型心肌梗死(STEMI)相关的潜在的分子标志物和治疗靶点。方法 从开源性基因表达数据库(GEO)获取STEMI相关芯片数据集GSE66360和GSE19339。利用R语言Limma包筛选差异表达基因(DEGs),使用韦恩图在线工具识别数据集共同的DEGs。利用clusterProfiler包对共同DEGs进行基因本体(GO)富集分析和京都基因与基因组百科全书(KEGG)信号通路分析;利用STRING数据库构建蛋白互作网络(PPI);采用Cytoscape(版本号3.9.1)软件对PPI进行可视化分析并筛选重要模块和关键基因。结果 通过对芯片数据集GSE66360和GSE19339进行综合分析,获得107个共同DEGs,包括81个表达上调基因和26个表达下调基因,其中,前10位的基因(IL-1β、ICAM1、FN1、vWF、PLAU、CXCL2、THBD、CCL20、CXCL3和CCRL2)均为表达上调基因;KEGG通路分析表明TNF信号通路、糖尿病并发症AGE-AGER信号通路和NF-κB信号通路是重要的通路。结论 监测白介素-1β等的动态变化,为预防STEMI发生提供了检测指标,同时也为STEMI的治疗提供了靶点和强有力的数据支持。Objective To screen potentially molecular markers and therapeutic targets associated with acute ST-segment elevation myocardial infarction(STEMI)by bioinformatics.Methods Obtaining STEMI related chip datasets GSE66360 and GSE19339 from the open source Gene Expression Omnibus(GEO).Used the R language Limma package to screen differentially expressed genes(DEGs),and the common DEGs were identified by the Venn diagram webtool.ClusterProfiler was used to Gene Ontology(GO)enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis for common DEGs.The STRING database was used to identify protein-protein interaction network(PPI).PPI visualization analyze and screening out of key genes were performed by using Cytoscape software(version 3.9.1).Results Through comprehensive analysis of the chip dataset GSE66360 and GSE19339,a total of 107 common DEGs were obtained,including 81 up-regulated genes and 26 down-regulated genes.Among them,the top 10 genes(IL-1β,ICAM1,FN1,vWF,PLAU,CXCL2,THBD,CCL20,CXCL3 and CCRL2)were all up-regulated genes.The KEGG pathway analysis demonstrated that TNF signaling pathway,AGE-RAGE signaling pathway in diabetic complications and NF-κB signaling pathway were the significant pathways.Conclusion Dynamic monitoring interleukin-1βlevel provides detection indicators for preventing the occurrence of STEMI,and also provides targets and strong data support for the treatment of STEMI.
