摘 要:目的:探讨骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)线粒体转移对促进内皮细胞(endothelial cells,ECs)迁移能力的影响。方法:采用线粒体提取试剂盒提取BMSCs线粒体,转移至同源同批次受体BMSCs中;采用Transwell法共培养BMSCs和ECs;采用划痕实验检测ECs迁移能力;采用Seahorse XFe-24分析检测细胞有氧代谢能力;采用ATP荧光检测试剂盒检测ATP浓度。结果:线粒体转移增强了与BMSCs共培养ECs的迁移能力(P<0.05),提升BMSCs和ECs能量代谢(P<0.05),抑制能量代谢后线粒体转移对BMSCs促进ECs迁移能力的作用被逆转(P<0.01)。结论:线粒体转移可能通过提升BMSCs和与其共培养ECs的能量代谢,促进ECs的迁移能力。Objective:To investigate the effect of mitochondrial transfer to bone marrow mesenchymal stem cells on promoting endothelial cell migration ability.Methods:Mitochondria were extracted from BMSCs using the mitoception method and transferred to homologous BMSCs from the same batch.The BMSCs and ECs were co-cultured using the Transwell method.Scratch assay was used to detect ECs migration ability.Seahorse XFe-24 analysis was used to detect cell aerobic metabolism.ATP luminescence assay kit was used to detect ATP concentration.Results:Mitochondrial transfer enhanced the migration ability of ECs co-cultured with BMSCs(P<0.05),and improved energy metabolism of BMSCs and ECs(P<0.05).The effect of mitochondrial transfer on promoting ECs migration ability by BMSCs was reversed when energy metabolism was inhibited(P<0.01).Conclusion:Mitochondrial transfer may promote ECs migration ability by increasing the energy metabolism of BMSCs and co-cultured ECs.
