摘 要:目的:通过建立坐骨神经损伤模型,观察周围神经损伤对骨髓间充质干细胞(BMSCs)生长增殖及成骨分化的影响。方法:取4周龄、健康、SPF级雄性SD大鼠20只,建立一侧下肢坐骨神经损伤模型,术后4周分别提取健侧(对照组)和术侧(实验组)骨髓间充质干细胞,进行体外培养增殖,采用流式细胞术检测BMSCs表面标志物,MTT实验检测2组细胞的活性;同时取P3代细胞行成骨诱导分化,并通过碱性磷酸酶(ALP)活性检测、茜素红染色对比2组细胞成骨分化能力。结果:流式细胞术检测提示2组细胞CD90阳性,CD11b阴性;MTT实验结果显示实验组BMSCs生长速度相对较慢,较迟进入平台期;成骨诱导分化后,实验组钙化结节、ALP活性的吸光度值均低于对照组,差异具有统计学意义(P<0.05)。结论:周围神经损伤可抑制BMSCs增殖及其向成骨细胞的分化。Objective To explore the effect of peripheral nerve injury on the proliferation and osteogenesis differentiation of bone mesenchymal stem cells(BMSCs)by establishing sciatic nerve injury model.Methods Twenty four-week-old,healthy,SPF grade male SD rats were selected to establish the sciatic nerve injury model of one side of the lower limb.Four weeks after operation,BMSCs were isolated from the healthy side(control group)and the operative side(experimental group),and cultured and proliferated in vitro.The surface markers of BMSCs were identified by flow cytometry,and the activity of cells in the two groups was detected by MTT assay;at the same time,P3 cells were taken for osteogenic differentiation,and the osteogenic differentiation ability of the two groups was compared by alizarin red staining and alkaline phosphatase(ALP)activity detection.Results The flow cytometry indicated that CD90 was positive and CD11b was negative in the two groups.MTT test showed that the growth rate of experimental group was lower than the control group.After BMSCs induced differentiation into osteoblasts,the absorbance value of calcified nodules and ALP activity in the experimental group were lower than those in the control group,and the difference was statistically significant(P<0.05).Conclusion Peripheral nerve injury are able to inhibit BMSCs proliferation and differentiation to osteoblasts.
