摘 要:【目的】采用高效液相色谱法对鲜黄花菜中的秋水仙碱进行检测。【方法】使用氨化二氯甲烷匀浆提取样品中的秋水仙碱,经C18固相萃取小柱净化,高效液相色谱仪-紫外检测器检测,外标法定量。【结果】秋水仙碱在0.02~1.00 mg/L范围内线性方程为y=42 404 x,相关系数r为0.999 5,在鲜黄花菜中添加0.1、0.5、2.0mg/L 3个水平的秋水仙碱,平均回收率为101%~106%,相对标准偏差为3.9%~5.2%,检出限为0.01 mg/kg。【结论】该方法精密度和准确性较高,能够在实际样品检测中应用。[Objective]The Colchicine in fresh daylily was detected by high performance liquid chromatography.[Methods]Colchicine was extracted from the samples with ammoniation of methanol by homogenate,purified by C18 solid phase extraction column,detected by high performance liquid chromatography-UV detector,and quantified by external standard method.[Results]The results showed that the linear equation of colchicine in the range of 0.02~1.00 mg/L was y=42404 x,and the correlation coefficient r was 0.9995.The average recoveries of colchicine in fresh daylily were 101%~106%,the relative standard deviations were 3.9%~5.2%at 0.1,0.5 and 2.0 mg/L level.The detection limit was 0.01 mg/kg.[Conclusion]The method has high precision and accuracy,which can be applied in sample detection.
