摘 要:目的构建8日龄小鼠急性心肌梗死(AMI)模型及模型标准化评估。方法取40只8日龄CD1小鼠,30只在体视显微镜下结扎心脏冠状动脉左前降支(LAD),建立AMI模型(AMI组),10只仅开胸不结扎冠状动脉(假手术组)。LAD结扎后1、3 d分别用TTC染色、TTC-伊文思蓝双染色法评估心肌缺血;LAD结扎后7、14、21 d通过心脏超声评估心脏结构和功能;结扎后4、7、14、21 d观察纤维瘢痕形成情况,Masson染色测量梗死纤维化面积。结果TTC染色和TTC-伊文思蓝双染色结果显示结扎线以下呈苍白色,说明成功造成心肌缺血。心脏超声结果显示:LAD结扎后7、14、21 d AMI组较假手术组左心室功能明显变差,左心室内径明显扩大,左室射血分数(LVEF)绝对值分别降低(37.64±4.30)%、(50.98±4.98)%和(46.49±5.49)%,左室短轴缩短率(LVFS)绝对值分别降低(31.06±2.99)%、(35.59±3.54)%和(30.40±4.07)%,左室舒张期内径(LVIDd)绝对值分别增加(0.69±0.30)mm、(1.25±0.47)mm和(1.35±0.41)mm,左室收缩期内径(LVIDs)绝对值分别增加(1.03±0.24)mm、(1.95±0.44)mm和(1.85±0.32)mm。Masson染色结果显示梗死区心肌组织逐渐被纤维瘢痕组织替代,结扎后4、7、14、21 d心脏纤维化面积分别达到(60.94±7.14)%、(67.85±11.4)%、(69.88±11.40)%和(75.79±7.67)%。结论本研究详细描述了8日龄小鼠AMI模型建立过程,通过不同方法评估了术后不同时间点心脏梗死面积﹑心脏结构﹑功能以及纤维化程度;为建立稳定可靠的8日龄小鼠AMI模型以及手术评估提供了参考数据。Objective To establish an 8-day-old mouse model of acute myocardial infarction(AMI)and its standardized evaluation.Methods A total of 408-dayold CD1 mice were used in this study.Left anterior descending coronary artery(LAD)was ligated in 30 mice under stereomicroscope to establish AMI model(AMI group),and thoracotomy was performed in 10 mice without coronary artery ligation(sham group).TTC staining and TTC-Evans blue double staining were used to evaluate myocardial ischemia one and three days after LAD ligation,respectively.Cardiac structure and function were evaluated by echocardiography on day 7,14 and 21 after LAD ligation.At 4,7,14 and 21 days after ligation,the formation of fibroscar was observed by gross observation,and the area of infarct fibrosis was measured by Masson staining.Results The results of staining showed a pale color below the ligation line,indicating that myocardial ischemia was successfully induced.The results of ultrasound showed that the cardiac structure and function of the AMI group were worse than that of the sham group at 7,14 and 21 d after LAD ligation.At 7,14 and 21 d after LAD ligation,compared with sham group,the left ventricular ejection fraction(LVEF)of AMI group decreased by(37.64±4.30)%,(50.98±4.98)%and(46.49±5.49)%respectively;left ventricular fractional shortening(LVFS)decreased by(31.06±2.99)%,(35.59±3.54)%and(30.40±4.07)%;left ventricular diastolic diameter(LVIDd)increased by(0.69±0.30)mm,(1.26±0.47)mm,and(1.35±0.41)mm;left ventricular systolic diameter(LVIDs)increased by(1.03±0.26)mm,(1.95±0.44)mm,and(1.85±0.32)mm.Masson staining showed that the myocardial tissue in the infarcted area was gradually replaced by fibrous scar tissue,and the cardiac fibrosis area reached(60.94±7.14)%,(67.85±11.4)%,(69.88±11.40)%and(75.79±7.67)%.Conclusion In this study,the process of establishment 8-day-old mouse AMI model was described in detail.The infarct size,cardiac structure,function and degree of fibrosis were evaluated at different time points by different methods afte
