摘  要:目的:探讨在瘢痕疙瘩成纤维细胞(Keloid fibroblasts,KFB)中叉头样转录因子3(Forkhead-like transcription factor 3,FOXP3)对真皮胶原蛋白沉积及纤维化的影响及相关机制。方法:免疫组化检测瘢痕疙瘩组织中I型胶原蛋白(Collagen type I,COL1)、Ⅲ型胶原蛋白(Collagen typeⅢ,COL3)、IL-6、TGF-β_(1)、FOXP3的表达。体外分离培养KFB,并采用腺病毒过表达FOXP3或给予TGF-β_(1)抑制剂处理。通过Western blot检测对照组、过表达组以及对照组和抑制剂组的COL1、COL3的表达,ELISA检测TGF-β_(1)、IL-6的表达,Masson染色观察胶原沉积情况。结果:与对照组相比,实验组中的COL1、COL3、IL-6、TGF-β_(1)蛋白水平升高,FOXP3蛋白水平降低。经过表达FOXP3或TGF-β_(1)抑制剂处理后,KFB的TGF-β_(1)和IL-6表达降低,胶原蛋白沉积减少。结论:在KFB中FOXP3可能通过抑制TGF-β_(1)的表达,减少组织纤维化和胶原蛋白沉积。Objective To investigate the effect of Forkhead-like transcription factor 3(FOXP3)on dermal collagen deposition and fibrosis in keloid fibroblasts(KFB)and related mechanisms.Methods Immunohistochemistry was performed to detect the expression of Collagen type I(COL1),Collagen type III(COL3),IL-6,TGF-β_(1),and FOXP3 in keloid tissues.KFB were isolated and cultured in vitro and treated with adenoviral overexpression of FOXP3 or given TGF-β_(1) inhibitors.The expression of COL1 and COL3 in control and overexpression groups as well as control and inhibitor groups were detected by Western blot,TGF-β_(1) and IL-6 by ELISA,and collagen deposition was observed by Masson staining.Results Compared with the control group,the levels of COL1,COL3,IL-6,and TGF-β_(1) proteins were increased and FOXP3 protein levels were decreased in the experimental group.After treatment with expression of FOXP3 or TGF-β_(1) inhibitor,TGF-β_(1) and IL-6 expression in KFB was decreased and collagen deposition was reduced.Conclusion In KFB FOXP3 may reduce tissue fibrosis and collagen deposition by inhibiting the expression of TGF-β_(1).
