摘  要:Heptahelical protein (HHP)转录因子家族在调节植物响应低温、盐害和激素处理等非生物胁迫中起着重要作用。生物钟能调控植物体的基因表达,光是调节植物生物钟,使植物与环境变化相适应的一个关键信号,然而对于HHP基因家族如何响应光信号的研究还鲜有报道。本研究利用生物信息学手段对大白菜BraHHP基因家族成员进行鉴定分析,并通过RT-qPCR方法,对BraHHP基因家族在不同光周期条件下的表达模式进行研究。本研究一共从大白菜基因组中鉴定出9个BraHHP基因,聚类在3个亚族中,BraHHP蛋白家族的氨基酸长度在323~387 aa,分子量在36.91~44.49 kD,所有蛋白成员均含有1个HlyⅢ保守结构域。另外,荧光定量结果表明,不同BraHHP基因在长日照(16 h光照/8 h黑暗)处理下的表达模式非常一致;在中日照(12 h光照/12 h黑暗)处理下,不同BraHHP基因之间的表达模式不尽相同;在短日照(8 h光照/16 h黑暗)处理下,除BraHHP2-2外,其他BraHHP基因的表达模式基本一致。本研究初步明晰了大白菜BraHHP基因家族对不同光周期的适应性反应,为进一步解析BraHHP基因家族的生物学功能提供了思路。Heptahelical protein(HHP)transcription factor family plays an important role in regulating plant resp-onse to abiotic stresses such as low temperature,salinity and hormone treatments.The biological clock can regulate the gene expression of plants,and light is a key signal for regulating the plant biological clock and making plants adapt to environmental change,however,there are few reports on how the HHP gene family responds to light sig-nals.In this study,the BraHHP gene family members of Chinese cabbage(Brass ica rapa L.ssp.pekinensis)were ide-ntified and analyzed by bioinformatics methods,and the expression patterns of BraHHP genes under different pho-toperiod conditions were studied by RT-qPCR.The results showed that 9 BraHHP genes were identified from the genome of Chinese cabbage and were clustered in three subfamilies,the amino acid length of BraHHP proteins ranged from 323 to 387 aa,the molecular weight ranged from 36.91 to 44.49 kD,and all protein members contained a Hly Ⅲ conserved domain.In addition,the results of fluorescence quantitative analysis showed that the expression patterns of BraHHP genes were the most consistent under long day condition(16 h light/8 h dark);The expression patterns of BraHHP genes were different under neutral day condition(12 h light/12 h dark);Under short day condition(8 h light/16 h dark),the expression patterns of BraHHP genes were basically the same except BraHHP2-2.This study preliminarily clarified the adaptive response of BraHHP gene family to different photoperiods in Chinese cabbage,and provided ideas for further analyzing the biological function of BraHHP gene family.
