摘  要:PIP基因家族是AQP基因家族中的一个亚家族,家族基因广泛参与运输水分及其他小分子、响应各种非生物胁迫、参与植物的开花与生长、气孔活动、种子萌发。本研究主要以蓖麻(大戟科)为材料,基于蓖麻基因组数据库,进行蓖麻PIP基因家族筛选及生物信息学分析。结果表明,共筛选出10个PIP基因成员,且不同蛋白质序列和特性都存在较大差异;基因结构分析结果表明,除了RcPIP10具有3个外显子,其余的都具有4个外显子,motif 1、motif 2、motif 5、motif 8是蓖麻PIP基因的特征基序;系统进化树结果表明,RcPIPs基因分成2个亚族且每个亚族均有5个基因,其中有2对基因在进化关系上成对出现,表现出较高的同源性;跨膜结构预测结果表明,蛋白RcPIP1~RcPIP9均有6个跨膜结构域,而RcPIP10仅有4个跨膜结构域,证实蓖麻家族PIPs大部分具有MIP超家族典型的跨膜螺旋特征;基因定位显示,10个基因不均匀的分布到9条染色体上,有8条染色体各分布1个RcPIP基因,仅有1条染色体分布2个RcPIPs基因。本研究结果不仅为大戟科植物和其他植物的PIP基因的功能分析和利用提供了有价值的信息,而且为基因家族扩展和进化研究提供了有益的参考。PIP gene family is a subfamily of AQP gene family.PIP genes are widely involved in water transport and other small molecules,response to various abiotic stresses,flowering and growth,stomatal activity and seed germination.In this study,the PIP gene family of castor(Euphorbiaceae)was screened and analyzed by bioinforma-tics based on castor genome database.The results showed that a total of 10 PIP gene members were screened,and different protein sequences and characteristics were quite different.The results of gene structure analysis showed that except RcPIP1O had three exons,the rest had four exons.Motif 1,motif 2,motif 5 and motif 8 were the characteristic motifs of Ricinus communis PIP gene.The results of phylogenetic tree showed that RcPIPs genes could be divided into two subgroups,each of which had five genes.Two pairs of genes appeared in pairs,showing high homology.The results of transmembrane structure prediction showed that RcPIP1~RcPIP9 had six transmembrane domains,while RcPIP10 had only four transmembrane domains,which confirmed that most of Ricinus communis family PIPs had typical transmembrane helix characteristics of MIP superfamily.Gene mapping showed that 10 genes were unevenly distributed on 9 chromosomes,8 chromosomes were distributed with one RcPIP gene,and only one chromosome was distributed with two Rc PIPs genes.These results not only provide valuable information for the functional analysis and utilization of PIP genes in Euphorbiaceae and other plants,but also provide useful references for the study of gene family expansion and evolution.
