摘 要:为了探讨海岛罗汉松(Podocarpus costalis)转录组功能基因信息,获取海岛罗汉松嫩叶叶色变化的相关基因,本研究对海岛罗汉松正常生长状态下的嫩叶进行转录组测序分析,去除低质量序列后得到52 189条Unigene,平均长度为1 609 bp,总长度为83 984 805 bp。52 189条Unigene注释到七大功能数据库,其中NR数据库中40 067个Unigene被注释到,注释率最高,占76.77%。在注释到的物种中,海岛罗汉松比对到的U nigene与北美云杉(Picea sitchensis)的相似度最高,共13 536条。通过5个时期的共同筛选,发现海岛罗汉松嫩叶有1 376个差异表达基因。1 376个差异表达基因通过GO分析注释到3个大类(生物过程,细胞组分,分子功能)的46个小类;KEGG分类可将1 376个差异表达基因分为5个大类19个小类。对与叶色变化相关的差异表达基因进行分析,发现与花青素合成相关的差异表达基因有9个,主要由苯丙烷类生物合成、类黄酮生物合成等机制影响海岛罗汉松嫩叶叶色变化;另外有3个差异表达基因涉及卟啉和叶绿素代谢途径。本研究结果为探究海岛罗汉松嫩叶叶色变化的分子机制提供了初步理论依据。In order to explore the functional gene information of the transcriptome of Podocarpus costalis and obtain the genes related to the change of the leaf color of Podocarpus costalis leaves,transcriptome sequencing analysis was conducted on young leaves of Podocarpus costalis under normal growth condition.After removing low-quality sequences,52189 Unigenes were obtained,with an average length of 1609 bp and a total length of 83984805 bp.52189 Unigenes were annotated to seven functional databases,among which 40067 Unigenes in the NR database were annotated,with the highest annotation rate,accounting for 76.77%.Podocarpus costalis showed the highest similarity to Picea sitchensis among the annotated species,with a total of 13536.It's found that 1376 differentially expressed genes were screened out in 5 periods.GO analysis showed that 1376 differentially ex-pressed genes annotated the molecular functions,the cellular components,biological processes and the 3 categories included 46 subclasses;KEGG classification can divide 1376 differentially expressed genes into 5 major categories and 19 subcategories.Analyzing the differentially expressed genes related to leaves color changes,it was found that there were 9 differentially expressed genes related to anthocyanin synthesis,which were mainly affected by mechanisms such as phenylpropane biosynthesis and flavonoid biosynthesis to the leaf color of Podocarpus costalis leaves.In addition,there were 3 differentially expressed genes involved in porphyrin and chlorophyll metabolic pathway.The results of this study provided a preliminary theoretical basis for exploring the molecular mechanism of young leaves color change in Podocarpus costalis.
