摘  要:为进一步探究CsKNAT3蛋白的功能与作用,本研究采用亚细胞定位,确定CsKNAT3能够定位于细胞核中作为转录因子发挥调节作用,同时采用原生质体瞬时转染确定CsKNAT3能够下调下游基因的表达,为转录抑制因子。研究通过酵母双杂交初步证实了CsKNAT3与CsATH1存在蛋白互作关系,双分子荧光互补进一步印证了这个蛋白互作,由于CsATH1是一个多效的植物发育调节因子,暗示CsKNAT3可能在植株发育过程中发挥重要调控作用。本研究对揭示KNOX家族蛋白调控植物形态建成分子机理具有重要意义。In order to further explore the function role of CsKNAT3 protein.In this study,the subcellular localization for CsKNAT3 was used to confirm that it can be located in the nucleus and play a regulatory role as a transcription factor.At the same time,the transient protoplast transfection was used to confirm that CsKNAT3 can negatively regulate the expression of downstream genes and act as a transcription repressor.The interaction of CsKNAT3 and CsATH1 was confirmed by yeast two hybrid system.The interaction between CsKNAT3 and CsATHI was further confirmed by BIFC.As CsATHI is a pleiotropic regulator of plant development,it imply thatCsKNAT3 may play an important role in plant development.The research is great significance to reveal the molecular mechanism of KNOX family proteins in regulating plant morphogenesis development.
