摘 要:本研究以‘苏椒1614’及其父母本为试验材料,利用SSR标记扩增条带多态性检测杂交种纯度。结果显示,从报道的29对核心SSR标记和课题组自主开发的3对InDel标记中筛选出19对双亲间具有多态性的标记。从中选取3对差异显著且带型简单的SSR标记Epms310、Epms331和Epms755对95个‘苏椒1614’单株进行纯度检测。3对引物检测结果完全一致,93个单株表现为父母本双亲互补条带为真实的F1杂交种,2个单株条带与母本材料相同为母本自交形成的杂株,种子纯度为97.9%。研究结果表明,‘苏椒1614’种子纯度达到97.9%,符合国家种子生产标准;筛选出的SSR标记可准确高效的检测杂交种纯度,可极大地节约杂交种纯度检测成本和时间。The purity of'Sujiao1614'hybrid seeds were evaluated by detecting bands polymorphism amplified by SSR markers of'Sujiao1614'and their parental inbred lines.The results showed 19 out of 29 primer pairs were with obvious differences between parents.Three SSR markers,Epms310,Epms755 and Epms331,with simple band patterns and the most significant differences,were selected to identify the purity of 95'Sujiao1614'plants.The identification results were completely consistent by 3 SSR markers.Ninety-three out of 95'Sujiao1614'plants with codominant bands of parental inbred lines were real F,hybrids.Two plants were amplified the same bands as the female parent.Therefore,the seed purity of'Sujiao1614'reached 97.9%,which was in line with the national seed production standard.The selected 3 SSR markers can be used to accurately and quickly identify the seed purity of'Sujiao1614'.
