摘 要:在棉花基因库日益贫乏的环境下,诱变育种技术成为发掘新基因以及种质资源创新的一个重要工具。本研究以新疆陆地棉品系‘新路早50’(XLZ50)的胚性愈伤组织细胞为实验材料,采用不同浓度的甲基磺酸乙酯(EMS)诱变剂进行诱变处理,用Na Cl作为选择剂,筛选出适宜棉花胚性细胞耐盐性鉴定的NaCl浓度,对筛选出的抗盐胚性细胞变异系,进行耐盐生理生化指标的检测。结果表明:导致XLZ50胚性细胞半致死率的EMS诱变处理浓度为0.6%;适宜XLZ50胚性细胞耐盐筛选的临界NaCl盐浓度为250 mmol/L;丙二醛含量和超氧化物歧化酶活性测定结果表明,耐盐突变细胞在生理指标上表现出与对照组较好的胁迫耐受能力。本研究初步探讨了细胞水平上获得EMS诱导突变耐盐植株的可能性。Under the circumstance that the gene pool of cotton is increasingly scarce,mutagenesis breeding tech-nology has become an important tool for germplasm resource innovation and new gene discovery.This study ap-plied somatic embryonic cells of Xinjiang upland cotton lines XLZ50 as experimental material,selected the proper methyl sulfonic acid ethyl ester(EMS)as the semi-lethal dose,and suitable NaCl concentration for screening EMS-mutagenesis embryonic cells as salt-tolerant selection agent,and then identified the surviving cells with the physiological items such as MDA content and SOD activities.The results illustrated that the semi-lethal dose for XLZ50 embryogenic cells is 0.6%EMS,and the critical NaCl concentration is 250 mmol/L for screening the salt-tolerant EMS-mutagenesis cells.The MDA content assay and the SOD activities result demonstrated that the surviving mutant cells display the better physiological items than the control group.Our study provided the the pos-sibility of obtaining salt-resistant cotton varieties at the cellular level with EMS mutagenesis.
