二倍体风信子‘Yellow Stone’和‘Red Pearl’核型比较分析

摘  要:经过多年的引种栽培,现代园艺风信子种质资源极其丰富。为找到有效鉴定风信子品种的方法,本研究采用常规压片法对两个二倍体风信子园艺品种‘Yellow Stone’和‘Red Pearl’进行根尖染色体核型分析,通过探究两品种的核型参数,以期为风信子种质资源鉴定提供可靠手段。结果显示,两个风信子染色体组成较为一致,均由中部(m)、近中部(sm)及近端(st)着丝粒染色体组成,均含有1对随体(sat)。其中,‘Yellow Stone’染色体相对长度变化范围为2.40%~11.05%,核型公式为2n=2x=16=6m+8sm+2sm (sat)。‘Red Pearl’染色体相对长度变化范围为2.73%~11.12%,核型公式为2n=2x=16=6m+8sm+2sm (sat)。两个品种的核型不对称系数分别为60.51%、59.59%,核型类型均为2C型。‘Yellow Stone’在染色体相对长度范围以及核不对称系数上均大于‘Red Pearl’。因此,综合核型类型与核不对称系数表明,‘Yellow Stone’的进化程度更高。Modern horticultural hyacinth germplasm resources are extremely rich after years of introduction and cultivation.In this study,to find an effective way to identify hyacinth cultivars,the karyotypes parameters of two diploid hyacinth horticultural cultivars'Yellow Stone'and'Red Pearl'were analyzed by routine root tip pressing method in order to probe into a reliable method for hyacinth germplasm resources identification.The findings indi-cated that the chromosome composition of two hyacinth cultivars are similar,of which are composed of cen-tromere chromosomes of median region(m),submedian region(sm)and subterminal region(st),and contain a pair of satellites respectively.The relative chromosome length of'Yellow Stone'ranged from 2.40%to 11.05%,and the karyotype formula was 2n=2x=16=6m+8sm+2sm(sat),while the counterpart ranged from 2.73%~11.12%,and the karyotype formula was the same.The karyotype asymmetry coefficients of two cultivars were 60.51%,59.59%,both the karyotype were 2C.'Yellow Stone'was larger than'Red Pearl'in the range of chromosome relative length and nuclear asymmetry coefficient.Therefore,the karyotype and nuclear asymmetry coefficient indicated that'Yel-low Stone'is of higher evolution.
