摘 要:该试验从大庆湿地水体中筛选出1株耐高温、耐高氨氮的异养硝化-好氧反硝化细菌。根据形态学特征及16S rRNA基因序列分析,鉴定并命名为Pseudomonas sp.LLM-5。采用单因素试验法,测定不同碳源、氨态氮浓度、C/N比和环境因素对该菌株脱氮效果的影响。结果发现,单因素试验中LLM-5菌株的最适脱氮条件为以丁二酸钠为碳源、C/N=16、盐度2%、初始pH 9.0。该菌株在最适温度(40℃)培养24 h后对氨氮、总氮、COD的去除率分别为87.38%、84.41%、79.90%。在初始氨氮浓度为1000 mg/L时,该菌对氨氮、总氮、COD的去除率分别达到94.34%、85.49%、82.55%,并在1200 mg/L时菌株仍能正常生长(OD_(600)=0.63)。菌株同时在以亚硝态氮与硝态氮为单一氮源(初始浓度为100 mg/L)时能将50%~60%的含氮物质去除。研究表明:LLM-5菌株具有优异的异养硝化性能、耐高温及高氨氮浓度,且具好氧反硝化性能,在处理高温、高浓度氨氮废水等领域具有广泛的应用前景。A heterotrophic nitrification and aerobic denitrification strain with high temperature and high ammonia nitrogen resistance was selected from Daqing wetland water.Based on morphological characteristics and 16S rRNA gene sequence analysis,it was identified and named as Pseudomonas sp.LLM-5.Single factor design was used to determine the effect of different carbon sources,ammonia nitrogen concentration,C/N ratio and environmental factors on the denitrification of LLM-5 strains.The results showed that the optimal nitrogen removal conditions of LLM-5 strain were sodium succinate selected as carbon source;C/N 16,salinity 2%,and the initial pH value 9.0.The removal rates of ammonia nitrogen,total nitrogen and COD were 87.38%,84.41%and 79.90%,respectively,after culture at the optimum temperature(40℃)for 24 h.When the initial ammonia nitrogen concentration was 1000 mg/L,the removal rates of ammonia nitrogen,total nitrogen and COD reached 94.34%,85.49%and 82.55%,respectively;and the strain could still grow normally with OD_(600)=0.63 at 1200 mg/L.The strain could remove 50%to 60%of the nitrogen-containing substances when nitrite and nitrate nitrogen were used as single nitrogen sources at initial concentration of 100 mg/L.This study showed that LLM-5 strain had excellent performance of heterotrophic nitrification,high temperature and high concentration of ammonia-nitrogen resistance,and the property of aerobic denitrification.It had a wide application prospect in the treatment of high temperature or/and highconcentration ammonia-nitrogen wastewater.
