摘 要:为了明确磷酸烯醇式丙酮酸羧化酶(phosphoenolpyruvate carboxylase,PEPC)序列在濒危岩牡丹属植物分子鉴定中的作用,本文研究了6种21份岩牡丹属植物,提取基因组DNA作为PCR扩增模板,对PEPC、matK和psbA-trnH基因片段进行扩增和测序。结果显示,岩牡丹属PEPC序列长度为679~1079 bp,matK序列长度为898 bp,psbA-trnH序列长度为307 bp。PEPC序列在岩牡丹属种间差异较大,可以准确区分岩牡丹属6种物种。基于PEPC序列构建的系统进化树优于matK和psbA-trnH组合序列构建的系统进化树。PEPC序列可作为岩牡丹属植物鉴定的DNA条形码。In order to examine the role of phosphoenolpyruvate carboxylase(PEPC)sequence in molecular identification of endangered plants of Ariocarpus Scheidw.,twenty-one samples of 6 species were studied,and gene segments of PEPC,matK and psbA-trnH were amplified and sequenced by PCR with DNAs extracted from the samples as templates.The results showed that the PEPC sequence length of Ariocarpus were 679-1079 bp,the matK sequence length were 898 bp,the psbA-trnH sequence length were307 bp.The PEPC sequence had great difference and could accurately distinguish the 6 species of Ariocarpus.The topology of phylogenetic tree which was constructed by the PEPC sequence was better than the tree of matK and psbA-trnH combined sequence.The PEPC sequence can be used as DNA barcode for identification of Ariocarpus.
