摘 要:目的:观察蒙药蓝刺头对骨质疏松(osteoporosis,OP)模型大鼠血清骨钙素(bone gla protein,BGP)调节、股骨近干骺端骨密度、股骨近干骺端形态改变、骨髓脂肪的影响。方法:去势复制绝经后骨质疏松症(postmenopausal osteoporosis,PMOP)大鼠模型63只,大鼠分为蒙药蓝刺头高、中、低剂量组及己烯雌酚组、强骨胶囊组、模型组、假手术组,每组9只,5天后通过宫颈涂片脱落细胞观察确定造模成功,90天后分别以蒙药蓝刺头高、中、低浓度,相应强骨胶囊、己烯雌酚分别灌胃90天;采集大鼠血清测定血清骨钙素(bone gla protein,BGP),剖取大鼠双侧股骨,左侧股骨近干骺端HE染色观察干骺端、骨髓形态、组织变化;显微CT右侧股骨骨密度(bone mineral density,BMD)指标分析。结果:血清BGP、BMD、股骨、骨髓脂肪改变模型组与假手术组相比,差别明显(P<0.01);适当剂量蒙药蓝刺头可有效降低血清BGP、提高BMD、改善股骨组织形态、抑制骨髓脂肪过快、过度表达(P<0.05);蒙药蓝刺头中剂量组同强骨胶囊组、己烯雌酚组效果相当(P>0.05)。结论:大鼠经去势造模,血清BGP明显增高;一定剂量蒙药蓝刺头可降低大鼠血清BGP,有明显抑制大鼠脂肪细胞密度、直径作用,还可抑制骨吸收、促进骨形成,改善骨组织形态,抗骨质破坏效果显著,提高BMD,为运用蒙医“补暖补精-清镇赫依-固骨质壮骨”理论防治OP明确部分作用机制,同时提供实验依据。Objective:To observe the effects of Mongolian medicine Lancitou on regulation of serum bone gla protein(BGP),bone mineral density(BMD),morphological changes of the proximal metaphysis of the femur,and marrow fat in OP rat models.Methods:All 63 PMOP rat models were duplicated after castration,rats were allocated to high,moderate and low doses groups of Lancitou,diethylstilbestrol(DES)group,Qianggu capsules group,the model group and sham operation group,nine rats in each group,cervical smear shedding cells were observed to confirm whether the modeling was successful in five days,the rats were drenched with high,moderate and low doses groups of Lancitou,the corresponding Qianggu capsules and diethylstilbestrol for 90 days respectively;rat serum was collected to detect BGP,after dissection of bilateral femurs from rats,HE staining of the proximal metaphysis of the left femur was used to observe the morphology and tissue changes of the metaphysis and bone marrow;microscopic CT was used to analyze BMD indicators in the right femur.Results:The difference was significant when the model group was compared with sham operation group in the changes of serum BGP,BMD,femur and the changes of bone marrow fat(P<0.01);the proper dose of Mongolian medicine Lancitou could effectively reduce the levels of serum BGP,raise BMD,improve femoral tissue morphology and inhibit rapid and overexpression of bone marrow fat(P<0.05);moderate dose group of Mongolian medicine Lancitou is equivalent to Qianggu capsules group and DES group(P>0.05).Conclusion:Serum BGP increases noticeably after castration and modeling in rats;a certain dose of Mongolian medicine Lancitou could lower serum BGP,showing the clear inhibitory effects on the density and diameter of rat adipocytes,and the significant effects of inhibiting bone resorption and promoting bone formation,improving bone tissue morphology and resisting bone destruction,as well as raising BMD,which could clarify some mechanisms of action and provide experimental reference for the prevention and t
