摘  要:目的探讨过表达硫化氢合成酶减轻去卵巢大鼠心肌缺血再灌注(IR)损伤的作用。方法选择雌性SD大鼠72只,其中32只随机分为假手术组、IR组、去卵巢组、联合组,每组8只。另外40只造模前进行去卵巢、尾静脉注射过表达硫化氢合成酶胱硫醚β合酶(CBS)或胱硫醚γ裂解酶(CSE)的腺相关病毒(AAV)。根据AAV尾静脉注射情况分为阴性假手术组、阴性IR组、阴性联合组、CBS联合组、CSE联合组,每组各8只。检测血清乳酸脱氢酶(LDH)、肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)水平以及细胞凋亡率。结果与IR组比较,联合组血清LDH、CK、CK-MB及心肌TNF-α、白细胞介素1β(IL-1β)、丙二醛、细胞凋亡率、裂解型半胱氨酸天冬氨酸蛋白酶3(Caspase-3)明显升高,超氧化物歧化酶(SOD)、LVEF、左心室短轴缩短率、CBS、CSE表达明显降低(P<0.05)。与阴性联合组比较,CBS联合组和CSE联合组血清LDH、CK、CK-MB水平明显降低[(207.65±20.74)U/L、(200.24±19.83)U/L vs(329.45±34.09)U/L,(201.32±24.05)U/L、(198.25±22.14)U/L vs(304.81±34.51)U/L,(56.33±5.72)U/L、(50.06±6.12)U/L vs(89.14±7.33)U/L,P<0.05],CBS联合组和CSE联合组心肌TNF-α、IL-1β、丙二醛、细胞凋亡率、裂解型Caspase-3明显降低,SOD含量明显升高(P<0.05)。结论去卵巢后心肌中硫化氢合成酶CBS和CSE表达降低,过表达CBS和CSE减轻去卵巢大鼠心肌IR损伤。Objective To explore the effect of overexpression of H2S synthase on myocardial IR injury in ovariectomized(OVX)rats.Methods A total of 72 female SD rats were randomly divided into sham operation group,IR group,OVX group,combination group,and negative sham group,negative IR group,negative combined group,CBS(H2S synthase cystathionine beta synthase)combined group,and CSE(cystathionine gamma lyase)combined group,with 8 rats in each group.Before modeling,the rats from the corresponding groups were ovariectomized,injected with adeno-associated virus(AAV)overexpressing CBS or CSE by tail vein.Serum levels of LDH,CK and CK-MB and apoptotic rate were detected.Results Compared with the IR group,the serum levels of LDH,CK and CK-MB,myocardial contents of TNF-α,IL-1βand MDA,apoptotic rate and expression level of cleaved Caspase-3 were significantly increased,while SOD content,LVEF and LVFS values,and CSE and CBS expression levels were obviously decreased in the combined group(P<0.05).The CBS combined group and CSE combined group had significantly decreased serum levels of LDH(207.65±20.74 U/L and 200.24±19.83 U/L vs 329.45±34.09 U/L),CK(201.32±24.05 U/L and 198.25±22.14 U/L vs 304.81±34.51 U/L)and CK-MB(56.33±5.72 U/L and 50.06±6.12 U/L vs 89.14±7.33 U/L),reduced myocardial contents of TNF-α,IL-1βand MDA,and lower apoptotic rate and expression level of cleaved Caspase-3,while elevated SOD contentwhen compared with the negative combined group(P<0.05).Conclusion The expression of H2S synthase CBS and CSE is decreased in the myocardium in rats after OVX,and overexpression of CBS and CSE can reduce myocardial IR injury in OVX rats.
