摘 要:目的:研究栀子苷对佐剂性关节炎(Adjuvant arthritis,AA)大鼠关节腔透析液中PGE_(2)含量的影响。方法:建立AA大鼠模型,采用关节腔肿胀度、关节炎症评分和大鼠全身表现评分等指标对大鼠模型进行评价。采集透析液,对关节腔药效学指标进行测定,观察关节腔中PGE_(2)水平随时间的变化情况,ELISA试剂盒检测大鼠血清中PGE_(2)蛋白含量,活性氧检测试剂盒检测活性氧变化。结果:与正常组比较,模型组足肿胀度有较显著增加(P<0.01)。同时,关节炎症评分及全身表现评分也有显著增加(P<0.01);与空白组比较,模型组中PGE_(2)浓度远远高于空白组,加入栀子苷药物后,PGE_(2)浓度有所降低;采用酶联免疫吸附法测定大鼠血清PGE_(2)含量时,与空白组比较,模型组血清PGE_(2)含量明显升高,差异有统计学意义(P<0.01);与模型组比较,药物组血清PGE_(2)含量均明显降低,差异有统计学意义(P<0.01),随着栀子苷药物剂量的变化,PGE_(2)含量逐渐下降;与模型组相比,栀子苷作用后的关节肿胀度有所降低,中高剂量时具有明显的降低效果(P<0.01)。在各组大鼠给药后关节炎症评分比较中,栀子苷作用后关节炎症评分下降,中剂量和高剂量作用效果明显,差异具有统计学意义(P<0.05);与模型组相比,栀子苷药物作用后全身评分降低,且差异具有统计学意义(P<0.05)。在ROS水平检测中,栀子苷作用后,活性氧水平有所下降,与对照组相比,具有显著性差异(P<0.01)。结论:栀子苷为栀子中活性最高的物质之一,实验研究对大鼠血清中PGE_(2)有一定的作用,且能够降低活性氧的水平,差异均具有统计学差异,表明栀子苷对关节炎大鼠有一定的治疗作用。Objective:To investigate the effect of gardenia glycosides on PGE_(2) content in articular dialysis solution in rats with adjuvant arthritis(AA).Methods:An AA rat model was established,and the rat model was evaluated by using the indicators of joint cavity swelling,joint inflammation score and rat overall performance score.Dialysate was collected,the pharmacodynamic indicators of the joint cavity were measured,the PGE_(2) level in the joint cavity was observed with time,the ELISA kit detected the PGE_(2) protein content in the serum of rats,and the reactive oxygen species detection kit detected the change of reactive oxygen species.Results:Compared with the normal group,the foot swelling degree in the model group was significantly increased(P<0.01).At the same time,the joint inflammation score and systemic performance score also increased significantly(P<0.01).Compared with the blank group,the PGE_(2) concentration in the model group was much higher than that in the blank group,and the PGE_(2) concentration decreased after the addition of genicoside drugs.When the serum PGE_(2) content of rats was determined by enzyme-linked immunosorbent method,the serum PGE_(2) content in the model group was significantly increased compared with the blank group,and the difference was statistically significant(P<0.01).Compared with the model group,the serum PGE_(2) content in the drug group was significantly reduced,with a statistically significant difference(P<0.01),and the PGE_(2) content gradually decreased with the change of the dose of gardenia.Compared with the model group,the degree of joint swelling after the action of gardenia was reduced,and there was a significant reduction effect at medium and high doses(P<0.01).In the comparison of post-administration articular scores of rats in each group,the articular score decreased after the action of gardeniain,and the effect of medium dose and high dose was obvious,and the difference was statistically significant(P<0.05).Compared with the model group,the systemic score after t
