油茶肉质果提取物的成分鉴定及抗结肠炎作用的研究

摘　　要：目的:研究油茶肉质果提取物(Camellia oleifera Abel fleshy fruit extract,CFFE)的成分及不同浓度CFFE抗葡聚糖硫酸钠(DSS)诱导的结肠炎作用。方法:使用液相色谱质谱联用技术(LC-MS)对CFFE的成分进行分析。将C57BL/6小鼠随机分为4组,先给药后造模。给药期间CFFE分别按高、低浓度给药,造模期间除对照组其余均进行3%DSS饮水处理。每天记录小鼠体重,处死小鼠时测量结肠长度,收集结肠组织分别通过RT-qPCR和ELISA方法测定肠道促炎症细胞因子的表达情况,HE染色检测炎症细胞浸润情况,使用Western-blot方法检测IκBα、p-IκBα、p38、p-p38、Erk、p-Erk、Jnk和p-Jnk蛋白表达情况。结果:CFFE含有多种成分,其中包括茶多酚、山柰酚和槲皮素等抗炎症分子。与对照组比较,模型组体重有明显下降、结肠长度缩短。与模型组比较,高浓度CFFE处理降低了IL-6和TNF-α的表达及炎症细胞浸润程度,且能显著抑制结肠组织的p-Jnk表达,但未影响p-IκBα、p-p38和p-Erk的表达。结论:CFFE含有多种抗炎症成分且对DSS诱导的结肠炎有预防作用,可能是通过抑制Jnk的活化来实现的。Objective:To investigate the components of Camellia oleifera Abel fleshy fruit extract(CFFE)and whether different concentrations of CFFE have anti-inflammatory effect on colitis caused by dextran sodium sulfate(DSS).Methods:CFFE components was analyzed by Liquid Chromatography mass spectrometry(LC-MS).C57BL/6 mice were randomly divided into 4 groups and administered first followed by modeling.CFFE was administered at high and low concentrations,and all groups except the control group were treated with 3%DSS water during the modeling period.The body weight of the mice was recorded daily,the length of the colon was measured at the time of execution,and the expression of intestinal pro-inflammatory cytokines was measured by RT-qPCR and ELISA,respectively,and the inflammatory cell infiltration was detected by H&E staining.The expression of IκBα、p-IκBα、p38、p-p38、Erk、p-Erk、Jnk and p-Jnk were detected by Western-blot.Results:CFFE had various components,including anti-inflammatory molecules such as Tea Polyphenol,Kaempferol and Guereetin.Compared with the control group,there was a significant decrease in body weight and shortening of colon length in the model group.Compared with the model group,high concentration of CFFE treatment reduced the expression of IL-6 and TNF-αand the degree of inflammatory cell infiltration,and significantly inhibited the expression of p-Jnk in colonic tissue,but it did not affect the expression of p-IκBα,p-p38 and p-Erk.Conclusion:It was found that CFFE had anti-inflammatory components and preventive effect on DSS-induced colitis,probably through the inhibition of Jnk activation.
