摘  要:【目的】应用蛋白组学技术解析镉(Cd)、硒(Se)胁迫对华贵栉孔扇贝(Chlamys nobilis)组织中蛋白表达的影响,为扇贝金属蓄积和耐受机制研究提供数据。【方法】将华贵栉孔扇贝置于分别含有Cd、Se、Cd+Se的海水中,采用非标记定量Label-free蛋白组学技术分析对照组和处理组扇贝消化腺的差异表达蛋白(differentially expressed proteins,DEPs),并对其进行GO富集、KEGG通路富集和蛋白质相互作用网络分析。【结果】CT组的闭壳肌和外套膜中的Cd质量分数显著高于鳃、性腺和消化腺(P<0.05)。除闭壳肌外,在镉、硒、Cd+Se胁迫下扇贝其他组织中的Cd或Se质量分数均显著增加(P<0.05)。Se+Cd胁迫下扇贝闭壳肌、外套膜和消化腺中的Se质量分数显著降低(P<0.05),说明扇贝中Se和Cd相互拮抗。共鉴定出887个DEPs;与对照组相比,Se组45个上调蛋白、9个下调蛋白,Cd组15个上调蛋白、10个下调蛋白,CdSe组13个上调蛋白、3个下调蛋白。生物信息学分析发现,Cd、Se胁迫的DEPs主要涉及参与细胞过程、代谢过程和生物调节,这些过程与KEGG通路分析所强调的信号通路有关。【结论】Cd、Se胁迫影响华贵栉孔扇贝消化腺多种蛋白的表达,导致相关代谢紊乱。【Objective】To analyze the effects of cadmium(Cd)and selenium(Se)stress on protein expression in Chlamys nobilis tissues by proteomics technology,and to provide data for the study of metal accumulation and tolerance mechanism of scallops.【Method】C.nobilis was placed in seawater containing Cd,Se,and Cd+Se,respectively,and then the differentially expressed proteins(DEPs)in the digestive glands of the control group and the treatment groups were analyzed by label-free proteomics technology,and the GO enrichment,KEGG pathway enrichment and protein-protein interaction network analysis were performed.【Result】The Cd content in adductor muscle and mantle of CT group was significantly higher than that in gill,gonad and digestive gland(P<0.05).The mass fraction of Cd or Se in other tissues except adductor muscle of scallops in metal treatment group increased significantly(P<0.05).The mass fraction of Se in adductor muscle,mantle and digestive gland decreased significantly(P<0.05)after scallops were placed in seawater containing Se and Cd,indicating that Se and Cd in scallops may antagonize each other.A total of 887 DEPs were identified.Compared with the control group,there were 45 up-regulated proteins and 9 down-regulated proteins in Se group,15 up-regulated proteins and 10 down-regulated proteins in Cd group,and 13 up-regulated proteins and 3 down-regulated proteins in CdSe group.Bioinformatics analysis revealed that the DEPs under Cd and Se stress were mainly involved in cellular processes,metabolic processes and bioregulation,which were related to the signal pathways emphasized by KEGG pathway analysis.【Conclusion】Cd and Se stress affect the expression of various proteins in the digestive gland of C.nobilis,resulting in related metabolic disorders.
