摘 要:试验旨在研究试验兔腹泻与粪便菌群之间的关系,通过16S rRNA基因测序对腹泻试验兔和健康试验兔的粪便菌群组成及丰度进行了研究。结果显示:共测得369888条有效序列,得到1738个OTUs,隶属于14个门、21个纲、53个目、97个科及217个属。腹泻晚期试验兔的粪便菌群α多样性指数显著降低(P<0.05),微生物丰富度显著下降(P<0.05)。腹泻粪便中厚壁菌门(Firmicutes)、梭状芽孢杆菌纲(Clostridia)、颤螺目(Oscillospirales)、毛螺目(Lachnospirales)、毛螺旋菌科(Lachnospiraceae)、瘤胃球菌科(Ruminococcaceae)及瘤胃球菌属(Ruminococcus)丰度明显下降,变形菌门(Proteobacteria)、γ变形杆菌纲(Gammaproteobacteria)、拟杆菌目(Bacteroidales)、肠杆菌科(Enterobacteriaceae)及埃希氏-志贺氏菌属(Escherichia-Shigella)丰度明显升高。腹泻兔和健康兔粪便中有309个OTUs存在显著差异(P<0.05),250个OTUs存在极显著差异(P<0.01)。LDA值大于2.4的菌群有38个,腹泻试验兔粪便中有5个显著差异菌群(LDA>3.6),分别为肠杆菌目(Enterobacterales)、肠杆菌科(Enterobacteriaceae)、埃希氏-志贺氏菌(Escherichia-Shigella)、Tissierellales菌及亨得特拉菌(Hungatella)。研究表明,腹泻兔粪便菌群多样性下降,菌群组成变化显著,肠杆菌目、肠杆菌科、埃希氏-志贺氏菌、Tissierellales菌及亨盖特拉菌为试验兔腹泻的生物标记。The purpose of the experiment was to study the relationship between diarrhea and fecal flora in rabbits.The composition and abundance of fecal flora in rabbits with diarrhea and healthy rabbits were studied by 16S rRNA gene sequencing.The results showed that a total of 369888 valid sequences were detected,and 1738 OTUs were obtained,belonging to 14 phyla,21 classes,53 orders,97 families and 217 genera.The fecal floraαdiversity index and microbial richness of rabbits with advanced diarrhea test were significantly decreased(P<0.05).The abundance of Firmicutes,Clostridia,Oscillospirales,Lachnospirales,Lachnospirae,Ruminococcaceae,and Ruminococcus in diarrhea feces decreased significantly,while the abundance of Proteobacteria,Gammaproteobacteria,Bacteroidales,Enterobacteriaceae and Escherichia-Shigella increased significantly.There was a significant difference in 309 OTUs between the feces of diarrhea rabbits and healthy rabbits(P<0.05),and a very significant difference in 250 OTUs(P<0.01).There were 38 bacterial communities with LDA values greater than 2.4,and there are 5 significantly different bacterial communities(LDA>3.6)in the feces of rabbits tested for diarrhea,namely Enterobacteriales,Enterobacteriaceae,Escherichia-Shigella,Tisierella,and Hungatella.The study indicated that the diversity of fecal flora of rabbits with diarrhea decreased and the composition of bacterial flora changed significantly.Thebiological markers of diarrhea were Enterobacteriaceae,Enterobacteriaceae,Escherichia-Shigella,Tissierellales and Hengetra.
