摘  要:目的通过高通量测序技术分析非哺乳期乳腺炎(non-puerperal mastitis,NPM)患者肠道菌群结构变化特征。方法2020年8月至2022年3月期间共纳入NPM组患者40例、健康对照组(healthy controls,HC)受试者30例,采集粪便样本后进行高通量测序,根据测序结果进行生物信息学分析。结果Alpha多样性分析显示,NPM组患者肠道菌群sobs、ace、chao、shannon指数显著低于HC组(P<0.05)。组间差异分析显示,NPM组与HC组相比,1个纲、5个目、14个科、28个属相对丰度存在显著差异。线性判别分析(linear discriminant analysis,LDA)显示在差异物种中1个纲、2个目、5个科和10个属组间差异影响显著(LDA值>3,P<0.5),其中10个菌属包括Faecalibacterium、Escherichia-Shigella在NPM组丰度富集,Anaerostipes、Streptococcus、Christensenellaceae_R-7_group、Alistipes、UCG-002、Ruminococcus_torques_group、Corynebacterium、Neisseria在HC组丰度富集,以上述10个菌属作为一个菌群组构建ROC曲线的AUC值为0.73,单个菌属中Anaerostipes、Ruminococcus_torques_group的AUC值分别为0.72、0.70,提示具有诊断意义。结论NPM患者肠道菌群丰富度、多样性显著低于HC组受试者,NPM患者与健康受试者肠道菌群组成存在显著差异。菌群组或单个菌属可作为区别NPM患者与健康人群的潜在生物标志物。Objective High-throughput sequencing was used to study on the changes of structural characteristics of gut microbiota in patients with non-puerperal mastitis(NPM).Methods A total of 40 NPM patients and 30 healthy controls(HC)were enrolled from Aug 2020 to Mar 2022.Stool samples were collected and then subjected to high-throughput sequencing.Bioinformatics analyses were performed based on the sequencing results.Results Analysis of Alpha diversity showed that the index of sobs,ace,chao and shannon of gut microbiota in NPM group were significantly lower than those in the healthy controls(HC)group(P<0.05).The difference analysis between groups showed that there were significant differences in the relative abundance of 1 class,5 orders,14 families and 28 genera between NPM group and HC group.Linear discriminant analysis(LDA)showed that among different species,1 class,2 orders,5 families and 10 genera had a significant impact on the difference between the groups(LDA>3,P<0.5),of which 10 genera including Faecalibacterium,Escherichia Shigella were enriched in the NPM group,and Anaerostipes,Streptococcus,Christensenellacea_R-7_group,Alistipes,UCG-002,Ruminococcus_torques_group,Corynebacterium and Neisseria were enriched in HC group.The area under the ROC curve(AUC)of the above 10 genera as a group of microbiota was 0.73.The AUC of Anaerostipes and Ruminococcus_torques_group as a single genus was 0.72 and 0.70,respectively,which indicated diagnostic significance.Conclusion The richness and diversity of gut microbiota in NPM patients was significantly lower than that in healthy control subjects.There are certain differences in the structure of the gut microbiota between the two groups.The microbiota group or single genus can be used as a potential biomarker to distinguish NPM patients from healthy people.
