提取时间和温度对高效液相色谱法测定维生素预混合饲料中维生素B_(2)含量的影响

摘　　要：试验旨在探讨提取温度和时间对维生素预混合饲料中维生素B_(2)测定结果的影响。采用两种不同载体类型的维生素预混合饲料(有机载体和无机载体),参照GB/T 14701—2019中高效液相色谱法进行操作,将提取温度分别设为80、90℃和100℃3个梯度,提取时间设为20、30、40 min和50 min 4个梯度,进行交叉试验,每个样品12个组,每组3个重复。结果显示:样品1(无机载体,主成分为沸石粉)中维生素B_(2)测定值随提取温度的升高不断提高,在100℃时的测定值显著高于其他温度时(P<0.05),而提取时间对测定值没有产生显著性影响(P>0.05);样品2(有机载体,主成分为玉米粉)在80℃、20 min条件下测定值偏低,其他条件下测定值没有显著差异(P>0.05)。结果表明,不同提取温度和提取时间能够影响不同载体维生素预混合饲料中维生素B_(2)的测定结果,其中以沸石粉为载体的维生素预混合饲料受提取温度影响较大,但在100℃、20 min的提取条件下均能够满足两种不同载体的提取要求。The purpose of the experiment was to study the effects of different extraction temperatures and extraction time on the determination of vitamin B_(2) in vitamin premix feed.Two kinds of vitamin premix feed,organic carrier and inorganic carrier were used in the experiment.The experiment process was conducted with reference of the high-performance liquid chromatography in GB/T 14701-2019,and the extraction temperature was set as three gradients at 80,90℃and 100℃,the extraction time was set as four gradients at 20,30,40 min,and 50 min.The cross test was conducted on 12 groups of each sample,with three replicates in each group.The results showed that the measured value of vitamin B_(2) in sample 1(inorganic carrier,which main component was zeolite powder)increased with the increase of extraction temperature,and the measured value at 100℃was significantly higher than other temperatures'values,while the extraction time had no significant difference on the measured value.The measured value of sample 2(organic carrier,which main component was corn flour)was lower than normal at 80℃and 20 min,and there was no significant difference in the measured value under other conditions(P>0.05).The experiment indicated that different extraction temperatures and extraction time could affect the determination results of vitamin B_(2) in vitamin premixes with different carriers.The vitamin premix used zeolite powder as carrier which was greatly affected by the extraction temperature,but it could meet the extraction requirements of the two different carriers under the extraction conditions of 100℃and 20 min.
