摘 要:目的:观察高强度间歇训练(HIIT)对心肌线粒体质量控制系统相关蛋白和糖代谢限速酶表达的影响,为阐明HIIT改善心肌能量代谢水平的机制提供理论依据。方法:将7周龄雄性C57BL/6J小鼠随机分为安静对照组(CON组,n=12)和高强度间歇训练组(HIIT组,n=12)。CON组小鼠不运动,HIIT组小鼠进行为期6周、每周5天的HIIT跑台训练。末次运动24 h后取小鼠心脏组织,剪取心脏组织左心室肌进行Western blot和RT-PCR实验。Western blot检测心肌线粒体质量控制系统相关蛋白和糖代谢限速酶的表达;RT-PCR检测心肌线粒体氧化磷酸化复合物Ⅰ~Ⅴ基因表达。结果:(1)HIIT对小鼠心肌线粒体质量控制系统相关蛋白的影响:与CON组相比,HIIT组线粒体生物发生蛋白过氧化物酶体增殖物激活受体-γ共激活因子1α(PGC-1α)、核呼吸因子1(NRF1)和线粒体转录因子A(TFAM)的表达增加(P<0.01,P<0.05,P<0.05),线粒体融合蛋白2(MFN2)的表达减少(P<0.05),线粒体融合蛋白视神经萎缩蛋白1(OPA1)的表达增加(P<0.05),分裂蛋白动力样相关蛋白1(DRP1)和分裂蛋白1(FIS1)的表达减少(P<0.05,P<0.01),自噬相关蛋白帕金森蛋白(PARKIN)的表达减少(P<0.01)。(2)HIIT对小鼠心肌线粒体氧化磷酸化复合物基因表达的影响:与CON组相比,HIIT组复合物Ⅰ(NDUFS8)、复合物Ⅱ(SDHb)、复合物Ⅳ(COX5b)和复合物Ⅴ(ATP5a1)的基因表达上调(P<0.01,P<0.05,P<0.01,P<0.05)。(3) HIIT对小鼠糖代谢限速酶的影响:与CON相比,HIIT组心肌糖酵解限速酶己糖激酶(HK)以及三羧酸循环限速酶柠檬酸合酶(CS)的表达增加(P<0.01,P<0.05)。结论:6周HIIT能够改善心肌线粒体质量控制系统相关蛋白的表达、上调线粒体氧化磷酸化复合物的基因表达水平以及增加糖代谢限速酶的蛋白表达,这些改变可能是HIIT改善机体心肌能量代谢水平的机制。Objective To observe the effect of high intensity interval training(HIIT)on the expression of proteins related to myocardial mitochondrial quality control system and rate-limiting enzymes of glucose metabolism,and to provide a theoretical basis for elucidating the mechanism of HIT improving myocardial energy metabolism.Methods Seven-week-old male C57BL/6J mice were randomly divided into a control group(CON group,n=12)and a high-intensity intermittent ttraining group(HIIT group,n=12).The mice of CON group did not exercise,while those of HITgroup underwent 6-week HIIT treadmill training for 5 days a week.Twenty-four hours after the last exercise,the left ventricular muscle of the heart was collected to evaluated the expression of proteins in myocardial mitochondrial quality control system and rate-limiting enzymess of glucose metabolism of using Western blot,as well as the gene expression of oxidative phosphorylation complex Ⅰ~Ⅴ in myocardial mitochondria using the RT-PCR.Results(1)Effects of HIIT on the myocardial mitochondrial quality control system related proteins:Compared with CON group,a significant increase was observed in the average expressions of the mitochondrial biogenetic protein peroxidase proliferator-activated receptor-coactivator 1α(PGC-1α),nuclear respiration factor 1(NRF1)and mitochondrial transcription factor A(TFAM),as well as the optic atrophy protein 1(OPA1)of HIIT group(P<0.01,P<0.05,P<0.05),but a significant decrease was found in the average expression of the mitochondrial fusion protein mitofusin 2(MFN2)(P<0.05),mitochondrial fission proteins dynamin related protein 1(DRP1)and fission 1(FIS1)(P<0.05,P<0.01),as well as the mitophagy related protein PARKIN(P<0.01).(2)Effects of HIIT on the genes expression of oxidative phosphorylation complex in the myocardial mitochondria of mice:Compared with CON group,gene expression of complex Ⅰ(NDUFS8),complexⅡ(SDHb),complexⅣ(COX5b)and complex Ⅴ(ATP5al)of HIIT group were significantly up-regulated(P<0.01,P<0.05,P<0.01,P<0.05).(3)Effects of
