摘  要:【目的】探讨中药秦巴硒菇提取物FA-2-b-β对慢性髓系白血病耐药细胞K562/ADR耐药性的逆转作用。【方法】采用CCK-8法检测细胞抑制率,流式细胞术检测细胞凋亡率、周期及细胞内药物蓄积情况,rt-PCR和Western blot法检测耐药及凋亡相关基因/蛋白表达,免疫荧光法观察K562/ADR细胞膜蛋白P-gp表达。【结果】不同浓度的FA-2-b-β对K562/ADR耐药细胞具有增殖抑制作用,其半数抑制浓度为(3.42±0.395)mg/mL,并可将K562细胞周期阻滞在G_(0)/G_(1)期,K562/ADR细胞阻滞于G_(2)/M期,且胞内荧光显示药物蓄积量增多。Western-blot及rt-PCR示FA-2-b-β能够明显下调耐药基因p65、MDR1的表达,上调基因p53、IκB-α、Bax的表达。【结论】秦巴硒菇提取物FA-2-b-β抑制K562/ADR细胞增殖并诱导其凋亡,其机制可能是通过调控NF-κB信号通路实现的。【Objective】To study the reversal of drug resistance in chronic myeloid leukemia resistant cells K562/ADR by Agaricus blazei extract FA-2-b-β.【Method】Cell inhibitory rate was determined by CCK-8 method,the apoptosis rate,cycle and intracellular drug accumulation by flow cytometry,the expression of apoptosis and drug resistance related genes by Western blot and RT-PCR,and the expression of P-gp protein by immunofluorescence.【Result】Different concentrations of FA-2-b-βhad proliferation inhibitory effects on K562/ADR drug-resistant cells,and its half-effective inhibition rate was(3.42±0.395 mg/mL).Western blot and qPCR results showed that FA-2-b-βcould significantly downregulate the expression of drug resistance genes p65 and MDR1,and up-regulate the expression of genes p53,IκB-αand Bax.【Conclusion】Agaricus blazei extract FA-2-b-βinhibited the proliferation and induced apoptosis of K562/ADR cells,and the mechanism may be achieved by regulating the NF-κB signaling pathway.
