益肾汤经结肠透析对慢性肾衰竭大鼠肠上皮细胞IRE1α、GRP78表达的影响

摘  要:目的:观察益肾汤经结肠透析对慢性肾衰竭(CRF)大鼠肠上皮细胞IRE-1α、GRP78表达的影响。方法:随机在60只健康雄性SD大鼠中抽取45只,腺嘌呤灌胃诱导建立CRF大鼠模型,其余15只为正常组。将造模成功大鼠随机分为CRF组、尿毒清结肠透析组和益肾汤结肠透析组,尿毒清结肠透析组予尿毒清结肠透析,益肾汤结肠透析组予益肾汤结肠透析,实验周期为4周。实验过程中记录大鼠一般状况、体重、血肌酐(Scr)、血尿素氮(BUN)的变化,4周末取肾组织、结肠组织行HE染色观察组织学改变,免疫组化检测各组结肠组织IRE1α、GRP78的表达及分布情况,实时荧光定量PCR法测定IRE1α、GRP78 mRNA表达情况。结果:(1)CRF组大鼠一般情况较正常组差,体重增长减慢(P<0.05),而尿毒清结肠透析组及益肾汤结肠透析组与CRF组相比有所改善,体重增加(P<0.05);(2)与正常组比较,CRF组Scr、BUN值升高(P<0.05),免疫组化示肠上皮细胞IRE1α、GRP78表达均明显上调(P<0.05),PCR结果显示GRP78、IRE1αmRNA相对表达量明显升高(P<0.05);(3)与CRF组比较,尿毒清结肠透析组及益肾汤结肠透析组Scr、BUN值降低(P<0.05),免疫组化示肠上皮细胞IRE1α、GRP78表达水平下调(P<0.05),PCR结果显示GRP78、IRE1αmRNA相对表达量显著降低(P<0.05),但两治疗组间表达差异无统计学意义(P>0.05)。结论:益肾汤经结肠透析可通过调节IRE1α、GRP78表达,抑制ERS保护肠黏膜屏障,从而改善CRF大鼠肾功能。Objective:To observe the expression of IRE-1αand GRP78 in intestinal epithelial cells of rats with chronic renal failure(CRF)by colonic Dialysis with Yishen Decoction.Methods:In order to produce a rat model of CRF,45 rats out of a total of 60 healthy male SD rats were chosen at random.The model group was then generated using the adenine gavage method.The normal group consisted of the remaining 15 rats in the experiment.After successful modeling,the rats in the model group were randomly divided into three groups:the CRF group,the Niaoduqing colon dialysis group,and the Yishen Decoction colon dialysis group.The Yishen Decoction colon dialysis group was given Yishen Decoction colon dialysis,while the Niaoduqing colon dialysis group was given Niaoduqing colon dialysis.The experimental period lasted for four weeks.During the course of the experiment,the general condition of the rats was evaluated,and the changes in the rats'body weight,serum creatinine(Scr),and blood urea nitrogen(BUN)levels were tracked and recorded.At the end of the fourth week,kidney tissue and colon tissue were collected for HE staining in order to observe histological changes.The expression and distribution of IRE1αand GRP78 in colon tissue of each group was detected and compared by immunohistochemical technique.Real-time fluorescence quantitative PCR was used to determine the mRNA expression of IRE1αand GRP78.Results:(1)The general condition of the rats in the CRF group was worse than that of the normal group,and the weight gain was slowed down(P<0.05).On the other hand,the general condition of the rats in the Niaoduqing colon dialysis group and the Yishen Decoction colon dialysis group improved in comparison with the CRF group,and their weight increased(P<0.05).(2)Compared with the the normal group,the levels of both Scr and BUN were found to be significantly higher in the CRF group(P<0.05).According to the findings of immunohistochemistry,the expression of IRE1αand GRP78 in intestinal epithelial cells belonging to the CRF group was signifi
