摘  要:目的:利用基因芯片分析寻找单侧输尿管梗阻(UUO)模型肾小管间质纤维化(TIF)的机制,并探究自噬在TIF中的变化。方法:选取GEO数据库中编号为GSE42303的基因芯片,提取其中假手术组与UUO组的肾组织基因表达谱进行分析。实验研究选取雄性SD大鼠,分成假手术组与模型组,每组各10只。在制备UUO模型14 d后取材,对梗阻肾组织进行光镜、免疫组化与透射电镜检测。结果:本研究共筛选出398个差异表达的基因,其中表达下调的有74个,上调表达的有324个。其中与自噬、炎症反应、氧化应激反应、小管离子转运、能量代谢以及胶原蛋白合成等生命活动相关的基因表达水平变化尤为显著。实验研究也进一步证实了梗阻肾组织中自噬激活与TIF的发生发展有密切联系。结论:TIF可能与自噬、小管离子转运障碍、能量代谢障碍、炎症反应、胶原合成以及腺苷通路活化有关,其中自噬可能是与TIF进展较为密切的关键机制。Objective:To investigate the mechanism of renal tubulointerstitial fibrosis(TIF)in the unilateral ureteral obstruction(UUO)model and to investigate the changes of autophagy in TIF.Methods:The gene chips numbered GSE42303 in GEO database were selected and the gene expression profiles of kidney tissues in the sham operation group and UUO group were extracted for analysis.Male SD rats were divided into sham operation group and model group,with 10 rats in each group.After 14 days of UUO model preparation,the obstructed kidney tissues were examined by light microscopy,immunohistochemistry,and transmission electron microscopy.Results:A total of 398 differentially expressed genes were screened in this study,among which 74 were down-regulated and 324 were up-regulated.The changes in gene expression levels related to autophagy,inflammation,oxidative stress,tubule ion transport,energy metabolism,and collagen synthesis were particularly significant.Experimental studies also further confirmed that autophagy activation in obstructed kidney tissue is closely related to the occurrence and development of TIF.Conclusion:TIF may be associated with autophagy,tubular ion transport disorder,energy metabolism disorder,inflammatory response,collagen synthesis,and adenosine pathway activation,among which autophagy may be a key mechanism and closely related to the development of TIF.
