摘  要:目的观察硅胶表面铜绿假单胞菌生物膜的形成,对比3种清洗剂对生物膜的清除能力。方法利用扫描电子显微镜观察经加片建模法构建的生物膜形态,采用结晶紫半定量和菌落计数定量法检测硅胶表面生物膜量。结果培养3d硅胶表面生物膜量明显高于d1和d5。清洗后扫描电子显微镜观察d3各组生物膜量明显减少但有残留。结晶紫半定量法证实Intercept清洗剂对培养3d的生物膜有极显著清除能力(0.19±0.05)(P<0.01);Zen清洗剂有显著清除能力(0.25±0.08)(P<0.05)。菌落计数定量法证实培养3d经Intercept和Zen清洗剂作用后菌落计数分别为(0.98±0.30)(P<0.01)、(2.79±1.15)×10^(1)(P<0.05)。鲁沃夫清洗剂对培养3d的生物膜清除无统计学意义。结论培养3d的铜绿假单胞菌生物膜量最多,黏附力最强。Intercept清洗剂对各个培养时间的生物膜均有清除能力,尤其对培养3d的成熟的生物膜也有较好的清除作用,Zen清洗剂效果次之。Objective To observe the formation of Pseudomonas aeruginosa biofilm on the surface of silica gel and compare the scavenging ability of three cleaning agents.Methods Scanning electron microscopy was used to observe the morphology of the biofilm constructed by adding slices,and the amount of biofilm on the surface of silica gel was measured by semi quantitative crystal violet and quantitative colony count.Results The amount of biofilm on the surface of silica gel cultured for 3 days was significantly higher than that on the 1st and 5th days.After cleaning,the amount of biofilm in each group was significantly reduced but there was residue on the 3rd day by scanning electron microscopy.The crystal violet semi quantitative method confirmed that Intercept detergent had a very significant scavenging ability(0.19±0.05)to the biofilm cultured for 3 days(P<0.01);Zen detergent had significant scavenging ability(0.25±0.08)(P<0.05).The quantitative method of colony count confirmed that the colony count after three days of culture by Intercept and Zen detergent was(0.98±0.30)(P<0.01),(2.79±1.15)(P<0.05),respectively.There was no statistical significance in the removal of bioflm by RUHOF detergent after 3 days of culture.Conclusion The biofilm of Pseudomonas aeruginosa cultured for 3 days has the largest amount and the strongest adhesion.Intercept detergent has a very significant scavenging effect on the biofilm on the surface of silica gel cultured for3days.
