摘 要:PBS1(AvrPphB susceptible 1)参与植物免疫反应,对植物免疫系统有着重要作用。为探究PBS1基因家族在大豆免疫系统中的作用机制,文章通过大豆基因组鉴定大豆PBS1基因家族成员,利用生物信息学软件及网站对大豆GmPBS1基因家族成员染色体位置、系统进化关系、保守motif、表达特征及启动子元件等进行分析。结果表明,大豆GmPBS1基因家族包含33个成员,不均等地分布在18条染色体上,分为Group A和Group B 2个亚族。亚细胞定位预测表明,大豆GmPBS1基因在细胞核、细胞质、细胞膜、线粒体和叶绿体中均有分布,主要定位于细胞核中。通过外显子-内含子可视化分析表明,同一亚族的基因结构不同,预示着GmPBS1在进化过程中可能出现了新的功能。进化树及共线性分析显示大豆GmPBS1基因家族存在4个复制基因对,分别为Glyma.07G136900和Glyma.07G195100、Glyma.11G147944和Glyma.12G074000、Glyma.14G025200和Glyma.02G289800、Glyma.03G176500和Glyma.03G253800。通过对大豆GmPBS1基因家族在不同组织中的表达量进行分析后发现,大豆GmPBS1基因在花和根组织发育中发挥重要作用。RT-qPCR结果显示,PBS1-1、PBS1-2和PBS1-3这3个基因在花、根和根瘤中均有着较高的表达量,推测大豆GmPBS1基因家族可能与花、根和根瘤的发育调控有关。大豆GmPBS1基因家族的启动子区元件多与抗逆和免疫相关,推测GmPBS1对植物免疫系统抵御逆境胁迫存在某种复杂作用机制,在大豆生长发育、开花结果、衰老死亡的整个生长过程中均发挥重要作用。通过蛋白网络互作预测发现,与PBS1蛋白互作的蛋白多为免疫相关蛋白,因此推测PBS1可能与植物免疫相关,在抗病方面发挥重要作用。PBS1(AvrPphB susceptible 1)played an important role in plant immune response.In order to explore the mechanism of PBS1 gene family in soybean immune system,the PBS1 gene family members were identified by soybean genome,and the chromosome positions,phylogenetic relationships,conserved motifs,expression characteristics and promoter elements of PBS1 gene family members were analyzed by bioinformatics software and website.The results showed that the GmPBS1 gene family contained thirty-three members,which were unequally distributed on eighteen chromosomes,and were divided into two subfamilies,Group A and Group B.Subcellular localization prediction indicated that GmPBS1 gene was distributed in the nucleus,cytoplasm,cell membrane,mitochondria and chloroplasts,and was mainly localized in the nucleus.The visualized analysis of exon-intron indicated that the different gene structures of the same subfamily indicated that GmPBS1 might had a new function during evolution.Phylogenetic tree and collinear analysis showed that there were four duplicate gene pairs in soybean GmPBS1 gene family.They were Glyma.07G136900 and Glyma.07G195100,Glyma.11G147944 and Glyma.12G074000,Glyma.14G025200 and Glyma.02G289800,Glyma.03G176500 and Glyma.03G253800.By analyzing the expression levels of soybean GmPBS1 gene family in different tissues,it was found that soybean GmPBS1 gene plays an important role in flower and root tissue development.RT-qPCR results showed that PBS1-1,PBS1-2 and PBS1-3 were highly expressed in flowers,roots and nodules,suggesting that the soybean GmPBS1 gene family might be related to the regulation of the development of flowers,roots and nodules.The elements of the promoter region of soybean GmPBS1 gene family were mainly related to stress resistance and immunity.It was speculated that GmPBS1 had some complex mechanism of action on plant immune system to resist stress,and played an important role in the whole growth process of soybean growth,development,flowering,fruiting,aging and death.Through the prediction of protei
