摘  要:旨在探讨多个营养转运相关基因调控区的多态性,并筛选影响肉鸡饲料转化率的关键变异位点。本研究采集了439只雄性黄羽肉鸡的血液DNA样本,利用PCR直接测序的方法在鸡群高、低饲料转化率的两尾样本组中(高FCR组>3.40,低FCR组<2.35,样本数均为12个)对3个单糖转运基因(GLUT2、GLUT 5和SGLT 1)、8个氨基酸转运基因(B^(0)AT1、B^(0+)AT、CAT1、CAT2、LAT1、y^(+)LAT1、y^(+)LAT 2和rBAT)和1个营养调控基因(FGF 1)上游1 kb内的多态性位点进行检测。依据两尾样本组中基因型频率的差异,筛选潜在影响饲料转化率的变异位点。采用PCR-RFLP技术在全群样本中对变异位点进行基因分型,并分析这些多态性位点与饲料转化率等性状的关联性。结果显示,本研究在12个基因的上游1 kb范围内共寻找到111个SNPs位点,其中有8个SNPs位点在两尾样本组中的基因型频率存在差异,分别为GLUT 2基因的-379A>G,SGLT 1基因的-490G>A,B 0AT 1基因的-639T>C,rBAT基因的-46G>A、-90G>T、-180T>C、-235G>A和FGF 1基因的-884T>C位点。在全群样本中对GLUT 2基因的-379A>G、B 0AT 1基因的-639T>C和FGF 1基因的-884T>C位点的基因分型和关联分析发现,B 0AT 1基因的-639T>C位点与饲料转化率存在显著相关(P<0.05),TC基因型个体的饲料转化率显著低于TT和CC基因型个体(P<0.05),表现出杂合子优势。FGF 1基因的-884T>C位点也与饲料转化率显著相关(P<0.05),CC和TT基因型个体的饲料转化率显著低于TC基因型个体(P<0.05),此外,GLUT 2基因的-379A>G与饲料转化率或生长性状均无显著相关。本研究分析了多个营养转运相关基因调控区的遗传变异,为将这些变异位点作为遗传标记用于提高黄羽肉鸡饲料转化率的分子选育提供了依据。The study aimed to investigate the polymorphism of regulatory regions of several nutrition transport-related genes and screen the key variations affecting the feed conversion ratio of broilers.The blood DNA samples of 439 male yellow-feather broilers were collected in this study,and the polymorphic loci within 1 kb upstream of three 3 monosaccharide transporter genes(GLUT2,GLUT 5 and SGLT 1),8 amino acid transporter genes(B ^(0)AT1,B ^(0+)AT,CAT1,CAT2,LAT1,y^(+)LAT1,y^(+)LAT 2 and rBAT)and 1 nutrient regulation gene(FGF 1)were detected in the two-tail sample group with high and low feed conversion ratio by PCR sequencing.According to the difference in genotype frequency in the two-tail sample groups,the genetic variations with potential influence on feed conversion ratio were screened.The PCR-RFLP method was used to genotype the polymorphic loci in the whole sample and the association between these polymorphic loci with feed conversion ratio and other traits was analyzed.The results showed that a total of 111 SNPs sites were found within 1 kb upstream of 12 genes in this study,among which 8 SNPs sites had different genotype frequencies in the two-tail sample groups,including-379A>G of GLUT 2,-490G>A of SGLT 1,-639T>C of B 0AT 1,-46G>A,-90G>T,-180T>C,-235G>A of rBAT,and-884T>C of FGF 1.Further whole population genotyping and association analysis showed that the-639T>C of B 0AT 1 was significantly associated with feed conversion ratio(P<0.05),individuals with TC genotype had significantly lower feed conversion ratio than those with TT and CC genotypes(P<0.05),showed a heterozygous advantage.The-884T>C of FGF 1 was also significantly associated with feed conversion ratio(P<0.05),and the individuals with CC and TT genotypes had significantly lower feed conversion ratio than those with the TC genotype(P<0.05).In addition,the-379A>G of GLUT 2 was not significantly associated with feed conversion or growth traits.In this study,the genetic variation of regulatory regions of several nutrient transport-related genes was an
